| Gallbladder carcinoma is the most common carcinoma of libiary tract and has high degree of malignancy. Because most of the gallbladder carcinoma is in the late stage ,the overall prognosis is poor. Gallbladder carcinoma has poor prognosis with 5%~13% actuarial survival at 5 years. Resection is the certain ways for patients with gallbladder carcinoma .Now the cause is not clear .Dysplasia of gallbladder mmucosa and gallbladder adenoma are recognized as precancerosis of gallbladder carcinoma .And some benign gallbladder diseases such as cholecystitis ,gallstones ,polypoid lesions of the gallbladder and anomalous junction of the pancreaticobiliary duct gradually may evolve into precancerosis of gallbladder carcinoma by the process of gallbladder carcinoma by the precess of pathophysiology ,then develop into gallbladder carcinoma .Ku70 protein is a ubiquitous protein in various in vivo. The current study found that Ku70 protein has many important and complex functions, including the Ku70 protein on the DNA double-strand breaks (DSBs) in the repair, Ku70 protein involved in regulating the function of DNA repair, Ku70 protein involved in immunoglobulin and T cell surface antigen Receptor V (D) J recombination function, Ku70 protein as a transcription factor regulating the expression of certain genes, Ku70 protein's role in nucleic acid metabolism, Ku70 protein structure with the role of telomere maintenance. Also on the distribution of Ku70 protein in human study found that Ku70 protein in several normal human organs and tissues and a variety of malignant tumors showed positive expression. And Ku70 protein expression in normal human tissues and its expression in malignant tissue there is a significant difference, Ku70 protein expression within the tumor tissue was significantly higher than in benign lesions and normal tissues. Present study aims to understand the Ku70 protein in gallbladder, gallbladder polyps and normal gallbladder tissue, discussed the occurrence of Ku70 protein in the gallbladder, the significance of the development process.Method of the present study by immunohistochemical method to detect Ku70 protein in 30 cases of gallbladder carcinoma, 20 cases of gallbladder polyps, 10 cases of normal gallbladder tissues. All patients with gallbladder cancer according to the UICC 2010 TNM staging edition, of which 2 cases stage IA, stage IB of the 14 cases, stage IIA in 8 cases, stage IIB 1 case, stage III in 3 cases, stage IV of 2 cases. Application of the experimental data, statistical software SPSS 12.0 was used for statistical analysis, statistical treatment using X2 test, when the P value of less than 0.05 was considered statistically significant.These results suggest that Ku70 expression in gallbladder, gallbladder polyps and normal gallbladder tissues have different levels of expression were 93.3%, 50%, 20%; Ku70 protein expression in gallbladder carcinoma of gallbladder polyps was significantly higher than And normal gallbladder tissue (P <0.05), and the later as the staging of gallbladder carcinoma, the worse the degree of differentiation is the expression of Ku70 protein was stronger; Ku70 protein in different histological types of carcinoma of gallbladder was no difference (P> 0.05). Treatment using X2 test, when the P value of less than 0.05 was considered statistically significant.In summary the following conclusions: Ku70 protein in normal gallbladder tissue, gallbladder polyps, and gallbladder cancer have different degrees of expression, but the Ku70 protein expression in gallbladder carcinoma was significantly higher than that of gallbladder polyps and normal gallbladder tissue. Through the Ku70 protein in different stages of gallbladder cancer, the positive rate of gallbladder cancer with analysis revealed that the later stages, the worse the degree of differentiation is the expression of Ku70 protein was stronger, suggesting that the expression of Ku70 protein And gallbladder cancer and malignant degree of correlation. Expression of Ku70 protein may be a common feature of certain malignant tumors. Increased expression of Ku70 protein may occur in the gallbladder, play an important role in the development process. |
