Objective: In this study, based on liver cancer model built in the SD rat, the contents of E-cd,β-catenin, MMP-7 in liver were measured at different stages of primary hepatic cancer to explore the relationship between the three elements and process of cancer which was developing.Methods: After 18 weeks of DENA gavage, liver samples were taken in the 2nd,4th,6th,8th,10th,12th,14th,16th,18thW respectively, then the contents of E-cd,β-catenin, MMP-7 were determined by immunohistochemistry and Western blotting. At last, all data were analyzed by SPSS software.Results:①According to the pathologic slices, the individuals in the medal groups appeared primary inflammatory reaction in the 2th-4th weeks, appeared severe inflammatory reaction in the 6th weeks, appeared adenoma hyperplasia in the 8th-10th weeks, appeared untypical adenoma hyperplasia in the 12th-14th weeks, appeared cholangiocarcinoma in the 16th-18th weeks.②immunohistochemistrial results: During the development of the rat liver cancer, in the modal group the E-cd positive rate decreases, and it was lower than the positive rate of the control signally during adenoma-hyperplasia-period (P<0.05); In the treat group, the positive rate of E-cd reduced more slowly than the treat group, but there was not a signal different between them (P>0.05). In the modal group theβ-catenin positive rate decreases, and it was lower than the positive rate of the control signally from inflammatory-reaction-period to cholangiocarcinoma-period (P<0.01); In the treat group,the positive rate ofβ-catenin was different from the treat group in the middle time, but there was not a signal different between them (P>0.05). The MMP-7 positive rate in the modal group increases, and it was higher than the positive rate of the control signally during untypical-adenoma-hyperplasia-period (P<0.05) and was higher than the positive rate of the control very signally during cholangiocarcinoma-period (P<0.01); In the treat group,the positive rate of MMP-7 induced more slowly than the treat group, but there was not a signal different between them (P>0.05).③Western blotting result:the content of E-cd in modal group was reducing gradually during the development of the rat liver cancer, and was lower than the content of control group in the cholangiocarcinoma-period (P<0.05); the content of E-cd in treat group was higher than the modal group, and was higher sigally than the content of modal group in the untypical-adenoma-hyperplasia-period (P<0.05). The content ofβ-catenin in modal group was was lower than the content of control group from the severe-inflammatory-reaction-period to cholangiocarcinoma-period very sigally (P<0.01); the content ofβ-catenin in treat group was lower than the modal group, and from the severe-inflammatory-reaction-period to cholangiocarcinoma-period the diffrent was signal (P<0.05), was very signal in the untypical-adenoma-hyperplasia-period (P<0.01).Conclusions: the content of E-cd andβ-catenin was reducing, MMP-7 was rising, and the place ofβ-catenin was changed. After the treat of Sophora Flavescens Ait and Se, the E-cd,β-catenin and MMP-7 were changed. Therefore, E-cd andβ-catenin, MMP-7, combined the interrelationships among them with their functions performed in the development of cancer could be demonstrated to become the important diagnosis markers in tumor pathogenesis and preventment. All of these produce new theory basic to the treatment of liver cancer.
