Background:More and more research prove that the gastric bypass operation on type 2 diabetic rats have an significant effection,surgical treatment on diabetes were accepted by more people, but we still do not know the mechanism of the surgical treatment.And we need to explore the true.Objective:To study the hypoglycemic effect of Gastric bypass(GBP) in streptozotocin (STZ)and high-glucose-high-fat diet-induced rat model of type 2 diabetes and evaluate the role of glucagon-like peptide-1, weight loss and decreased food intake after operation.Methods:twenty-four male SD diabetic rats induced by STZ and high-glucose-high-fat diet were randomly allocated into four groups:GBP(group O), shame operation(group S), food restriction (group F) and controls (group C). Plasma glucose concentration, insulin, glucagon-like peptide 1 (GLP-1) were measured respectively at fasting and postprandial condition before and the 1st week,2nd week,3rd week,4th week,8th week after GBP in all rats, weight and food intake were measured simultaneity.Results:Three weeks after operation in the GBP rats, mean fasting plasma glucose concentration declined from (16.7±3.7)mmol/L to (13.1±3.4)mmol/L and postprandial plasma glucose from (31.7±6.1)mmol/L to (17.2±3.3)mmol/L (P<0.05), fasting plasma insulin concentration rose from (28.55±8.26)IU/L to (46.37±10.32)IU/L and±postprandial plasma insulin from (30.62±8.88)IU/L to (51.23±11.34)IU/L (P<0.01), fasting plasma GLP-1 concentration rose from(7.11±2.21)pmol/L to (25.25±7.21)pmol/L (P<0.01) and postprandial plasma GLP-1 from (42.11±11.25)pmol/L to (97.72±30.12)pmol/L (P<0.01), and marked change happened four weeks after GBP.These results were not seen in the group S despite similar same postoperative weight gain profile. GBP resulted in better glycemic control than greater weight loss from food restriction.Conclusion:GBP can markedly reduce plasma glucose in type 2 diabetic rats, and hypoglycemic effect may owe to more GLP-1 secreted after GBP. The effects are not secondarily to weight loss or lower food intake after operation.
