ObjectiveTo investigate the expressions of the inhibitor genes of apoptosis of Survivin,PCNA, P53,Caspase-9 in the epithelial ovarian tumor,analyzing expression differences between benign and malignant,the realationship with clinical and pathological features,and expres sion related Survivin genes and P53,PCNA,Caspase-9 protein,To explore the potential rol es of Survivin in the development and progress of the epithelial ovarian cancers.MethodsUsing S-P immunohistochmistry to detect the expressions of Survivin,PCNA,P53, Caspase-9 in different epithelial ovarian tissue;Using molecular techniques in situ hybridi zation to detect the expressions of Survivin mRNA.Analysizing two different methods to detect the expressions consistency between Survivin mRNA and Survivin protein,and the expressions consistency between Survivin and P53 protein;Analysizing protein expression rate comparation in Survivin,PCNA,P53,Caspase-9,and the relationship with clinical feat- ures.Results1.The positive rates of Survivin mRNA in 38 cases of EOC were 71.05%,in 11 cases of benign tumor were 18.18%,in 9 cases of normal ovary tissues were 11.11%.The expression of Survivin mRNA in EOC and in the benign tumors were significant different- ces as well as in EOC and in normal ovary tissues(P<0.05,P<0.05).The expression of Sur vivin mRNA was no significant differences in the benign tumors and in normal ovary tissues(P>0.05).The expression of Survivin mRNA in EOC is related with the histological grade,FIGO staging, lymphnode metastasis and cancerousascits(P<0.05),but the expression is no related with the pathological type(P>0.05).2.The positive rates of Survivin protein in 38 cases of EOC were 84.21%,in 11 cases of benign tumor were 27.27%,in 9 cases of normal ovary tissues were 11.11%.The expres- sion of Survivin in EOC and in the benign tumors were significant differences as well as in EOC and in normal ovary tissues(P<0.05,P<0.05).The expression of Survivin was no significant differences in the benign tumors and in normal ovary tissues(P>0.05). The expression of Survivin in EOC is related with the histological grade,FIGO staging, lymph- node metastasis and cancerousascits(P<0.05),but the expression is no related with the pathological type(P>0.05).3.The positive rates of p53 in 38 cases of EOC were 78.95%,in 11 cases of benign tumor were 27.27%,in 9 cases of normal ovary tissues were 22.22%.The expression of p53 in EOC and in the benign tumors were significant differences as well as in EOC and in normal ovary tissues(P<0.05,P<0.05).The expression of p53 was no significant differences in the benign tumors and in normal ovary tissues(P>0.05).The expression of p53 in EOC is related with the histological grade,and cancerousascits(P<0.05),but the expression is no related with the pathological type,FIGO staging, lymphnode metastasis(P>0.05).4.The positive rates of PCNA in 38 cases of EOC were 39.47%, in 11 cases of benign tumor were 18.18%,in 9 cases of normal ovary tissues were 11.11%.The expression of PCNA in EOC and in the benign tumors were significant differences as well as in EOC and in normal ovary tissues(P<0.05,P<0.05).The expression of PCNA was no significant differ ences in the benign tumors and in normal ovary tissues(P>0.05).The expression of PCNA in EOC is related with the histological grade,FIGO staging,lymphnode metastasis(P<0.05), but the expression is no related with the pathological type and cancerousascits(P>0.05).5.The positive rates of Caspase-9 in 38 cases of EOC were 39.47%,in 11 cases of benign tumor were 81.82%,in 9 cases of normal ovary tissues were 88.89%.The expression of Caspase-9 in EOC and in the benign tumors were significant differences as well as in EOC and in normal ovary tissues(P<0.05,P<0.05).The expression of Caspase-9 was no significant differences in the benign tumors and in normal ovary tissues(P>0.05).The ex pression of Caspase-9 in EOC is related with the lymphnode metastasis(P<0.05),but the expression is no related with the histological grade,FIGO staging, pathological type and cancerousascits(P>0.05).6.The expressions of Survivin and p53 had posititive correlation in the same sample (rs=0.350,P<0.05).7.The expressions of Survivin and PCNA had posititive correlation in the same sample(rs=0.521,P<0.05),The expressions of Survivin and Caspase-9 had negative correl- ation in the same sample(rs=-0.323,P<0.05);The expressions of p53 and PCNA had positi- tive correlation in the same sample(rs=0.504,P<0.05),The expressions of p53 and Caspase -9 had negative correlation in the same sample(rs=-0.431,P<0.05). 8.The expressions of Survivin mRNA and Survivin protein had consistency in the same sample,in the ovrian benigh tumor(k=0.500,P=0.025),but in the ovrian cancer(k= 0.333,P=0.032).Conclusion1.The expression of Survivin mRNA and Survivin protein were significantly increase- ed in ovarian cancer,and have consistency each other.associated with the degree of differ- entiation,clinical stage,lymph node metastasis and ascites.2.The expression of Survivin,PCNA,P53 protein in ovarian cancer was significantly increased,but expression of Caspase-9 protern decreased significantly,indicating that there may play an important role in development,invasion and metastasis of ovarian cancer.The four proteins and clinicopathological features have some of relevance,their expression lev- el of can serve as useful indicators of malignancy in detecting ovarian cancer.3.The expression of Survivin has positively correlated respectively with PCNA and p53,but has negatively correlated with Caspase-9,the expression of P53 has positively correlated with PCNA,but has negatively correlated with Caspase-9.Survivin and P53 have synergy in the process of the pathogenesis of ovarian cancer.4.Detection the four gene of ovarian cancer has great value to estimate assessment of the biological behavior,it can help to estimate prognosis of patients,provide the basis for clinical treatment,and maybe become a new molecular target therapy site in treatment of ovarian cancer. |
