1,ObjectiveBezoar ginseng capsule is according to the "Yiqi detoxification"of the basic principles, contains a certain percentage the Vitro cultivated calculus bovis and ginseng. The use of serum pharmacology method can be study the effect of the drug-containing serum on HepG2 about human hepatoma cell cycle and apoptosis status and to explore its mechanism.2,MethodsBy a certain proportion to the original crude drug of bezoar ginseng capsule made decoction, rats were given a certain dose of decoction and were divided into drug-containing serum group and blank control group. Application of serum pharmacology method, the two groups were diluted to different concentrations of serum(10%,20%,50%), bring up HepG2. Then use the growth curve assay, fluorescence staining fluorescence microscopy, flow cytometry double staining method, test and compare the effect on HepG2 about human hepatoma cell cycle and apoptosis status,3,Results3.1. The Influence on human hepatoma cell HepG2 cells growth. The detection of the growth curve from the cell growth of the experimental results, found that after 72h culture, the cell survival rate:Card declined to 0,20%, platinum group containing serum group dropped to 0.05%,50% serum containing group dropped to 33.33%; cultured 72h, blank serum and blank control group, decreased survival rate is not obvious.3.2. The Influence on human hepatoma cell HepG2 cells cycle. To sum up Experimentâ… and Experimentâ…¢, serum containing different concentrations were able to act on human hepatoma cells HepG2, different segments of the cell cycle have persistent effects, and mechanism of action of carboplatin is different, the study also need to be further explored.3.3. The Influence on human hepatoma cell HepG2 cells apoptosis.Fluorescence microscopy observation from the results, was found to contain serum group were able to make fluorescence intensity, the cell shrinkage, nuclear condensation, formation of apoptotic bodies, and promote the apoptosis of HepG2 human hepatoma cells.From the flow cytometry results, containing different concentrations found in serum can promote apoptosis of human hepatoma cells, compared with the control group had significant differences compared with the control serum group was significantly different.4,Conclusions 4.1. Drug-containing serum can affect the human hepatoma cell HepG2 cells growth.4.2. Drug-containing serum can affect the human hepatoma cell HepG2 cells cycle.4.3. Drug-containing serum can affect human hepatoma cell HepG2 cells apoptosis.4.4. Drug-containing serum inhibits the growth of human hepatoma cell HepG2 and promoting the apoptosis in human hepatoma cell HepG2, and the serum concentration and incubation time was associated. The most evident effect is the 20% drug-containing serum group.
