The Change Of Neutrophils' Function And Activity In Acute Infection And Other Surgical Clinical Research

ObjectiveIn this program, we design an experiment to research the relationship between neutrophils and necrotic cells. By obtaining some important information, we focus the research of acute appendicitis and get the similar data which could direct us the treatment options from patients suffered from acute infection Anti-infectiveResultsBy flow cytometry, we initial proof that necrotic cells could attract neutrophils.Neutrophil which swallowed necrosis would die. The dead mode was various by different time and the quantity of necrotic cells.According to the patients of acute appendicitis, pure neutrophil could enhance the quality of the experiment. And separated liquid that isolated neutrophils (just 80%) was lower than the manetic microbeads (94.5%). So we isolated neutrophils by magnetic microbeads in the experiment.We found that by infection (acute appendicitis), neutrophils' founction was significantly strengthen and the survival time was prolong. Neutrophils which cultured by 72-hour, infected stage of acute appendicitis patients, recovery stage of acute apendicitis and healthy volunteers. The last two groups of neutrophils were about 70% cells had died. But the infected group of neutrophils was only 6.5% died. This period may be associated infection with bone marrow neutrophils which product new cells. We further analyze the function of neutrophils in various stages of all groups. We found that infected group (2h,9.27%) was swallow quicker than other two groups (4h,6%).Infected neutrophils phagocytosis beads mainly died in apoptosis. While the recovery group and healthy group occurred necrosis. For example, in the 4th hours, we examined the rate of apoptosis and necrosis in each group:the infection group's apoptosis rate was 81.45%, while the apoptosis rate of recovery and the healthy groups was only 30.34% and the necrosis rate was 63.5%.Conclusion1,the necrotic cells could attract neutrophils.2,Neutrophil which swallowed necrosis would die.
