Objective Our previous research[1] found in part of the lung injury caused obstruction in the rat model, Dachengqi Decoction(DD) can reduce pulmonary edema, interstitial thickening, bleeding and inflammatory cells infiltrating etc of pathology, Confirm this model rats DD has protective effect.To observe the effects of the medicated serum containing DD on expressions of Caveolin-1(CAV-1),eNOS,NF-КB p65 and TLR-4 in model of lung damaged, DD medicated further explores the protective effects of this model serum mechanism.Methods Dachengqi Decoction and Medicated serum were prepared. Then HBEs were divided into randomly the following 7 groups :normal serum group, LPS group, the low-dose medicated serum group, the middle-dose medicated serum group, the high-dose medicated serum group, Positive control group, Negative control group),expression of CAV-1,eNOS,NF-КB and TLR-4 mRNA assayed by immunocytochemistry Real-time PCR and Western blot .Result DD medicated MTT results showed that of LPS exciting HBE serum has protective effect. The inhibitory action of the high medicated serun group was more obviously than the low medicated serun group(p<0.01).Contrasted to LPS group, the normal rabbit serum group was decreased in the level of CAV-1,eNOS,NF-КB p65 and TLR-4 protein(p<0.01),CAV-1,eNOS,NF-КB p65 and TLR-4 in different medicated serun groups(20%,10%, 5%)was contrasted to LPS group decline . A basal level of CAV-1,eNOS,NF-КB p65 and TLR-4 in HBE were detected in Normal serum group, after 24hours of the LPS stimulation, compared with the normal serum group, the expression of CAV-1,eNOS,NF-КB p65 and TLR-4 mRNA increased dramatically in the LPS group(p<0.01), the different dose medicated serun can suppress the expression of CAV-1,eNOS,NF-КB p65 and TLR-4 mRNA in HBE exposed to LPS.Conclusion Dachengqi Decoction medicated serum inhibit on expression of CAV-1,eNOS,NF-КB p65 and TLR- 4 in HBE stimulated by LPS, and This may indicate LPS DD medicated HBE damage to stimulate serum has protective effect.
