Objective: Cardiovascular diseases (CVD) are the leading cause of illness and death around the world, and prevention and treatment of CVD is becoming the focus in health relative area. Epidemiological studies have indicated a strong inverse association between grape wines intake and CVD. Resveratrol, a polyphenolic phytochemical found in wine, has been speculated to play vital role for this phenomenon. With its antioxidant, anti-inflammatory, anti-platelet aggregation, anti-proliferation and anti- carcinogenic potential, resveratrol have been studied, especially the effects on anti-carcinogenic potential and prevention of CVD. It was reported that resveratrol can ameliorate endothelial function with underlining mechanism illegible. Endothelium is not only a monolayer of cells lining the lumen of the vascular beds, but also a pivotal modulation organ of vascular homeostasis. We investigated the effects of resveratrol on eNOS and Cav-1 gene expression to explore mechanism of endothelial function improvement.Methods: In vitro, serial subcultivation bovine artery endothelial cells (BAECs), passage 5 to 15 were treated with 0, 0.01, 0.1, 1 and 10μmol/L resveratrol for 20h respectively. After extracting protein from treated cells, the eNOS protein, p-eNOS (ser1177) protein, Cav-1 protein, p-Cav-1 protein levels were detected by Surface Plasmon Resonance Biomolecular Interaction Analysis (SPR) and Cav-1 protein level was detected by Western Blot method.Result: (1) p-eNOS protein level was enhanced after BAECs had been treated with 0, 0.01, 0.1, 1 and 10μmol/L resveratrol for 20h. The maximal up-regulation of p-eNOS was observed with 1μmol/L group. As for eNOS expression, the maximal up-regulation was observed in 0.1μmol/L group. (2) Cav-1 protein expression was down-regulated after BAECs had been treated with 0, 0.01, 0.1, 1 and 10μmol/L resveratrol for 20h. The maximal down regulation of Cav-1 was observed with 1μmol/L group. Conclusions: resveratrol can up-regulation the gene expression of eNOS and enhanced phosphorylation of eNOS (ser1177), down-regulate gene expression of Cav-1 dose-dependently.
