Objective To investigate the effect of caveolin-1(CAV-1) gene transfected on the expression of toll-like receptor4(TLR-4)induced by lipopolysaccharide(LPS) in human bronchial epithelial cells (HBE) and the intervention effect of emodin on the expression of CAV-1and TLR-4.Methods This experiment was divided into two parts:the first part of the experiment,given different concentrations emodin intervention to LPS-induced HBE cells,with the establishment of the normal control group, the LPS alone intervention group, the positive control group (atorvastatin) and the negative control group (solvent DMSO, dimethyl sulfoxide),the protein of HBE cells from different intervention groups were collected to measure the expression of CAV-1by Western blot; the second part of the experiment,building the CAV1-EGFP recombinant plasmid and transfecting the HBE cells transiently, with establishment of the normal control group and empty plasmid group, detecting and comparing the CAV-1expression of three groups by Western blot,otherwise,LPS was administered into the cell culture medium at48h post-transfection, Western blot detect and compare the TLR-4expression of HBE before and after transfection;and emodin was given into the LPS-induced HBE cells after transfection,the expression of TLR-4was assayed by Western blot.Result the first part of the experiment, compared with the normal control group,CAV-1 protein expression of HBE cells were significantly increased after stimulated by LPS(P <0.05);both the middle-does emodin and the high-does emodin can suppress the expression of CAV-1in LPS-induced HBE cells(P<0.05);the second part of the experiment,compared with the normal control group and the empty plasmid transfected group, CAV-1expression of the CAV-1-EGFP recombinant plasmid group was significantly increase (P<0.05),but the empty plasmid group compared with the control group, the CAV-1expression was no statistically significant (P>0.05);compared with control groups,CAV-1over-expression could raise LPS-induced HBE cells the expression of TLR-4but emodin can inhibit this effect.Conclusion Emodin can inhibit the expression of CAV-1in LPS-induced HBE cells;CAV-1gene transfection can up-regulate the expression of TLR-4in LPS-induced HBE cells,but emodin can inhibit this effect.
