Objective:Dry eye is a common, multiple ocular surface disease .The patient usuallypresents dryness , foreign body sensation and photophobia,and so on.The reasons ofincidence is various, new study shows that inflammation plays an important role in dryeye.Cyclosporine is a fungal metabolite, which is an immunosuppressive agent.Domesticand international research shows that it works by inhibiting the aggregation ofinflammatory cells and release of inflammatory factors,reducing lacrimal gland andconjunctiva's lymphocytes infiltration , inhibiting the apoptosis.of ocular tissues such aslacrimal gland and conjunctival goblet cells. This study aims to discuss the clinical efficacyof cyclosporine for dry eye and safety assessment.Methods: Select 20 patients who were diagnosed as dry eye in Union Hospital of TongjiMedical College from 2000,7 to 2011, 4.40 eyes in all,and be divided into control group andstudy group.Control group were given artificial tear three times a day and cyclosporinesolvent twice a day.Study group were instructed to instill artificial tear three times a day andcyclosporine eye drops twice a day.shimmer I test,tear break up time,ocular surface stainingwere assessed at basline , 1week,and 1,2,3months.in addition,vision and self-symptomswere asked every time. ResultResults: (1) Study group and control group have no significant differences in observedindicators at basline(p>0.05). (2)study group were superior to control group in threeindicators at 1m,2m,3m(BUT:P<0.02,P<0.01,P<0.01; SIT:P<0.05,P<0.01,P<0.01;ocularsurface staining:P<0.05,P<0.02,P<0.01),But no significant difference at 1w(P>0.05).(3)cyclosporine group,but not artificial tear group improved BUT ,SIT.,ocular surfacestaining indicators at 1m,2m,3m(BUT:P<0.01,P<0.01,P<0.01; SIT:P<0.01,P<0.01,P<0.01).(3)two groups improved ocular surface staining scores(P<0.01,VS basline).study groupfavored at 1w,1m,2m,3m(P<0.01,P<0.01,P<0.01,P<0.01),control group effects at1m,2m,3m(P<0.01,P<0.01,P<0.01).Conclusions: 0.05%Cyclosporine eye drops can effectively relieved the signs andsymptoms of dry eye, increased tear secretion, and improved the ocular surfaceenvironment Objective: To learn the effects of 0.05% CsA eye drops on conjunctival goblet cells of dry eye models.Methods: Select 20 SD rats, both male and female.5 as the A group, no treatments.15 be made into dry eye models by alkali burn.Each group have 5 rats and be divided into B,C,D three groups.the B group Without treatments after alkali burn.The C group be given artificial tears three times a day and the D group be given cyclosporine eye drops twice a day.All rats be Killed after 1 month and took the conjuctival to do paraffin section and HE stainning,observed the changes of goblet cells.Results: (1) The A group was the normal SD rats' conjunctiva paraffin sections.A large number of goblet cells present Between the pseudostratified epithelium. (2)The B group,goblet cells almost disappeared and conjunctiva epithelium thinned.(3)The C group was treated with artificial tears.There were few goblet cells.(4)The D group was treated with cyclosporine eye drops after alkali burn.more goblet cells presented and conjunctival epithelial thickened.Conclusions Cyclosporine eye drops can significantly promoted goblet cells' recovery of dry eye model by alkali burn.Better than artificial tears.
