Objective Preliminary discussion the effect of estrogen applied to human sperm cryopreservation, to provide a more effective cryoprotective medium (CPM). Methods Collected sixty human sperm from Reproductive center of Ningxia Medical Univerdity (Normal semen group:20 cases, Oligoastenozoospermic azoospermia group:40 cases). Each semen will be divided into five copies, one copy is used control group, the other four copies are used experimental group. Conventional semen analysis, viability, motility, morphology and spermatozoa hypo-osmotic swelling rate (HOSR) were used to evaluate the cryo- protective effect of different CPMs, glycerol, glycerol-eygyolk-sodium(GYC), glycerol+ estro -gen, GYC+estrogen on human spermatozoa. In the experimental process, the estrogen concentration we used was 500ng/L, the sperm movement parameters was best after thawing. Results During the procedure of cryotective and thawing, different cryoprotective medium has different protective effect on the spermatozoa, the viability, motility, morphology and hypo-osmotic swelling rate decreased after thawing, there was statistically significant(P<0.01). Compare the glycerol group, the viability, motility, morphology and hypoosmotic swelling rate of GYC group has significant differences (P<0.01). Compared with glycerol and GYC group, there were significantly differences in the viability, motility of glycerol+estrogen and GYC+estrogen(P<0.01). But there were no significant differences in the morphology and hypo-osmotic swelling rate(P>0.05). Conclusions (1) The viability, motility, morphology and hypo-osmotic swelling rate decreased after thawing, vitrification inflicts obious damage on sperm. (2) The viability, motility, morphology and hypo-osmotic swelling rate of GYC group significantly higher than the glycerol group, the protection of mixed cryoprotective medium superior to the glycerol group. (3) The viability, motility of the glycerol+estrogen and GYC+estrogen significantly higher than the glycerol and GYC group, prompt that the cryoprotectant containing estrogen can increase the function of cryopreserved sperm. Estrogen is a feasible sperm cryoprotectant.
