Objective To explore the correlation between the Rho/Rock signal pathway in human sperm and the anti-freezing protection of sperm,to provide a theoretical basis for clinically improving the sperm cryopreservation recovery rate,the development of highly effective semen cryoprotectants,and optimizing assisted reproduction technology(ART).Methods To take 20 healthy volunteers' fresh semen and divide each semen into 3groups: basic data group,normal reagent control group and Rho / Rock pathway inhibitor group,and fast freeze the semen of the last two groups,and test the separately before freezing and after resuscitation sperm resuscitation parameters(viability,morphology,sperm membrane integrity rate,acrosome enzyme activity,DNA fragments,mitochondrial membrane potential,acrosome response)of each group,statistical analysis,to explore the correlation between Rho/Rock signaling pathway in sperm and sperm anti-freezing protection and its clinical significance.Results Rho/Rock pathway inhibitor group sperm motility [(56.90 ± 7.16)% vs(51.45 ± 7.00)%,P = 0.002],sperm survival rate((61.33 ± 4.33)% vs(52.96 ± 5.22)%),P =0.001],sperm membrane integrity rate [(68.14 ± 3.76)% vs(59.79 ± 5.60)%,P = 0.001],percentage of normal morphology sperm [(7.52 ± 1.41)% vs(4.90 ± 1.20)%,P = 0.001],sperm DFI [(18.70 ± 4.38)% vs(27.10 ± 6.53)%,P = 0.001],sperm mitochondrial membrane potential[(63.26 ± 3.25)% vs(56.06 ± 3.80)%,P = 0.001],the indicators are significantly better than those of the normal refrigerant group.There was no significant difference in acrosomal enzyme activity between the inhibitor group and the normal cryotherapy group [(97.25 ± 11.91)%vs(97.35 ± 9.58)%,P >0.05].Immunofluorescence staining showed that RHOa / ROCK proteinwas widely expressed in the sperm head and neck.Conclusion Rho/Rock signaling pathway inhibitors have a certain role in sperm freezing injury,which can significantly improve the ability of sperm to resist freezing injury.
