Objective:To compare the effect of PROH and DMSO with EG and DMSO as Cryoprotective Agent on morphology and function of ovarian tissue after Slow Freezing. And to compare the effect of VEGF,VE,As and VEGF,VE with no medicament anti-injury of treatment on morphology and function of freezed ovarian tissue after Autotrans-plantation.Methods:The fifty-four rats were randomly assigned into nine groups with six rats each. Ovariectomized operatie were done from the first group to the eighth. PROH and DMSO as cryoprotective agent freezed the first,third,fifth groups ovarian tissue; EG and DMSO as cryoprotective agent freezed the second,fourth,sixth groups ovarian tissue; The seventh group was autotransplantion the fresh ovarian tissue. The eighth group was as the ovariectomized group and the ninth was as the normal control group. Slow Freeze the ovrian tissue of the first group to the sixth which had been in full of cryoprotective agent vessel; and put the vessel into liquid nitrogen for two weeks; then autotransplante. VEGF,VE and As apply anti-injury of treatment on the first and the second groups. VEGF and VE apply anti-injury of treatment on the third and the fourth groups. None apply on the fifth and the sixth group. At two month after implantation blood were collected to detect the level of estradiol (E) and ovarian tissue were reclaimed to analyze their morphological changes.Results:Compare the first group to the second,the third to fourth,the fifth to the sixth the level of estradiol,the morphological and the quantity of normal cells of ovarian tissue after apply the method of slow freezing,two cryoprotective agents and VEGF,VE,As or none anti-injury of treatment, and take the T-test. t1.2=2.395,P 1. 2<0.05; t3.4=2.714, P3.4<0.05; t5.6=2.480, P5.6<0.05. It means anti-injury of treatment do or not the PROH and DMSO as cryoprotective agent groups the level of estradiol higher than the EG and DMSO as cryoprotective agent groups. Apply VEGF,VE,As *and VEGF,VE** the function of ovarian tissue are better than no medicament groups((P*<0.05;P**<0.05). But apply VEGF,VE,As groups no better than VEGF,VE groups (P>0.05); The change of the ovarian tissue morphological the VEGF,VE,As groups and VEGF,VE groups are better than no medicament groups. But there is no statistics divergence.Discussions:PROH and DMSO as cryoprotective agent is better than EG and DMSO on reducing injure the tissue of ovary and restoring the function of ovary by using the slow freezing.So PROH and DMSO is the better cryoprotective agent. Apply VEGF,VE,As and VEGF,VE anti-injury of treatment are more useful to restore the ovarian tissue morphological and function than no medicament groups Apply VEGF,VE and As no better than only apply VEGF and VE.
