Objective:This study investigated the variation of the expression of BimEL mRNA and P-BimEL protein in in the cochlear tissues of the SD rats and analyzed its possible role in the pathogenesis of the ototoxicity induced by cisplatin. Material and Methods: 80 female Sprague-Dawley rats,randomly divided into five groups. In the control group (n=16), only 10 ml/ kg 0.9%NaCl was injected;Animals were killed at 2 days (n=4), at 4 days (n=4), at 7 days (n=4) and at 30 days (n=4). Cisplatin 5mg /kg in 1.0 ml normal solution (0.9%NaCl) was injected intraperitoneally. Animals was killed as follows: (1) (n=16) rats were killed 2days after CDDP administration,(2) (2) (n=16) rats were killed 4 days after CDDP administration, (3) (n=16) rats were killed 7 days after CDDP treatment and (4) (n=16)rats were killed 30 days after CDDP injection. BimEL mRNA expression was detected by Real-time polymerase chain reaction (Real-time PCR) and P-BimEL protein expression was evaluated by Western Blot.Results: It was found that the expression level of BimEL mRNA was elevated significantly in 2 day group compared with the control and other groups(p<0.05). 4 day group, 7day group, 30day group and NS group compared with each other did not show any significant difference(P>0.05) Western Blot: It was found that the expression level of P-BimEL was elevated significantly in 2day group , 4 day group, 7day group compared with the control and 30 day groups. (p<0.05). 7 day groups compared with 2 day group and 4 day group was elevated significantly(P<0.05). other groups did not show any significant difference(P>0.05).Conclusions: BimEL may be involved in the pathogenesis of the ototoxicity induced by cisplatin and regulated by transcription and post-translation.
