ObjectiveTo investigate the effect of intestinal electrical stimulation (IES) on postprandial blood glucose in healthy volunteers and its possible mechanism.Mathods1. Six healthy volunteers were enrolled in a 2-session study (real and sham sessions). The order of the two sessions was randomized and 3 days apart.2. The subjects were required to take a normal, balanced diet the week before taking the test. Volunteers were fasted for at least 8 to 12 hours before having the test.3. All volunteers were incubated with a nasal duodenal feeding tube with 2 ring electrodes into the duodenum .The location of the electrodes were checked by X-ray to confirm the right position in the duodenum. In this session,we drew blood at different time point and measured gastric emptying half time(T1/2) with ultra sound.4. The postprandial blood glucose at different time point 2 hours after the meal, were measured with real or sham IES.5. Symptoms of dyspepsia,which including abdominal pain,appetite,bloating, fullness,nausea and vomiting were scored every 30 minutes in these two sessions . 6. The level of insulin was determined by radioimmunoassay in these two sessions.Results1. The postprandial blood glucose at different time point 90min after the meal in real IES session were significantly lower than those in the sham session.2. The serum insulin at 60min,90 min and 120min after the meal in real IES session were significantly lower than those in the sham session.3. The T1/2 of gastric emptying was 40.67±5.72 min in the real session, which was significantly higher than that in the sham session (27.33±4.03 min, p <0.05).4. The dyspeptic symptoms score in real session was significantly higher than that in sham session (p<0.05).Conclusions1. IES can decrease the postprandial glucose level, which contribute to its effect on delaying gastric emptying ,inhibiting the absorption of glucose and other unknown mechanisms.2. IES may have potential applications for the treatment of diabetes.
