Objectives To investigate the effect of recombinant human interleukin-17 (rhIL-17) on vascular endothelial growth factor (VEGF) expression in human NSCLC cell lines A549 and H460 cultured in vitro.Methods First, A549 and H460 cell lines were cultured in complete medium without or with rhIL-17 at different concentrations (0.1, 1, 10, 100 ng/ml). Then after 48h, 72h of cell culture, MTT assay was performed to evaluate cell proliferation. After 24h co-culture with rhIL-17, semi-quantitative RT-PCR was done to detect the VEGF mRNA in them. The expression of VEGF protein was also measured following 48h co-culture with rhIL-17 by enzyme linked immunosorbent assay (ELISA).Results Treated with different concentration rhIL-17 for 48h or 72h, no significant difference was observed between control and rhIL-17-treated groups in A549 cell proliferation. For H460 cell proliferation, there is no statistical difference among groups after 48h treatment, while the proliferation was obviously inhibited in groups with 10 ng/ml and 100ng/ml rhIL-17 after 72h. No statistical difference was detected on the expression of VEGF mRNA and protein in A549 and H460 cells which were treated with varying concentration of rhIL-17.Conclusions Exogenous rhIL-17 at concentrations (0.1, 1, 10, 100 ng/ml ) had no direct effect on the proliferation of A549 cell in vitro, but could inhibited proliferation of H460 cell at the concentrations of 10 ng/ml and 100 ng/ml rhIL-17. Neither the expression of VEGF mRNA nor protein was induced by rhIL-17 in A549 and H460 cells.
