Alzheimer's disease is a progressive brain neurodegenerative disorder. The deposition of (3-amyloid protein causes the activition of microglia and releasing of inflammatory factors that induces the chronic inflammatory reaction, which may be the most important pathological mechanism of Alzheimer's disease pathogenesis. Rescarchs indicate thatβ-amyloid protein can activate microglia and express inflammatory factors (IL-1β, IL-6, TNF-α, TGF-βM, etc) in high level.ObjectiveTo explore the effects on microglia of oligomeric forms ofβ-amyloid protein(Apo). To further analyze the influence of Kadsura Pepper Stem Extract on the upper progress.Methods1. Primary cultivation of microglia taken from newborn rats(within three days), purified and identified.2.Use ethanol to extrat Piper kadsura Stem. The extract was filtered and concentrated under reduced pressure; blended with DMSO in the proportion of 4:1; add NS; then micro filtrated;3.Purified microglia was inoculated into 96-well plates by the same density and divided into six groups:①blank group;②aβo stimulated microglia for 48h;③aβo+KPSE for 48h;④aβo+DMSO for 48h.Detect levels of TNF-a and TGF-β1 in cell's supernatant liquid by enzyme-linked immunosorbent assay(ELISA).Resultsl.Aβo can stimulate microglia to express inflammatory fators compared with blank group:aβo groups can be detected with more inflammatory fators;2.KPSE suppresses the expression of inflammatory factors of microglia:the level of IF in KPSE group is lower than the control group.ConclusionsAβo can activate micoglia and causes the release of IF; KPSE can suppress the release of IF.
