AimsGastric cancer is one of the most common malignant tumors in the world.In China. 17 million people died from gastric cancer each year,which accout for.25% of cases died from cancer And there were about 40 million patients with gastric cancer each year. so the cancer is indeed a serious disease that threats to people's health. Gastric cancer occurs in different age groups,.most in 40 to 60 years. The number of male with gastric cancer have two times as many as famele.In recent years the age of people with gastric cancer became younger gradually. Because of the symptoms of gastric cancer and precancerous lesions are unspesific, early diagnosis is difficult. In fact, we can find only 5% to 10% of early stomach cancer, it is not ideal.Therefore, it is extremely important to study the growth, adhesion, invasion and metastasis of gastric cancer cell, to guide clinical treatment. P-FAK is a tyrosine kinase, which plays an important regulatory role in growth, development, migration of cancer cell, and its expression is high in tumor tissues. And Paxillin is protein kinase containing tyrosine and combine with a cancer gene.It is involved in biological functions of regulation of tumor cell with phosphorylation of FAK and other proteins. The phosphorylation of FAK and Paxillin are related to development, invasion and metastasis of many tumor cells.But we do not understood fully the mechanism of them.The reports of expression with joint detection in the stomach cancer are not be finded.The present study detects the expression of The phosphorylation of FAK and Paxillin in normal gastric tissues and gastric adenocarcinoma, to evaluate the function of the phosphorylation of FAK and Paxillin in the occurrence, development, invasion and metastasis process of gastric adenocarcinoma, and to explore their relationship with the occurrence invasion and metastasis of gastric cancer cells,Materials and methods:85 patients with 63 males and 22 females who had undergone gastric adenocarcinoma in department of Pathology, the First Affiliated Hospital of Zhengzhou university from January 2008 to December 2009, were enrolled in this study. Gastric adenocarcinoma had been confirmed by histopathological diagnosis in all tissue samples All patients were not received preoperative chemotherapy,radiotherapy and immunotherapy before operation.The range of ages were 32~80 years.The average age were 55.5 years.TNM staging:Ⅰperiod of 15 cases;Ⅱperiod of 20 cases,Ⅲperiod of 22 cases,Ⅳperiod of 28 cases;10 well-differentiated and 40 moderately and 35 poorly differentiated; 40 below subserosal layer and 45 above subserosal layer; 50 with lymph node metastasis and 35 without lymph node metastasis. In addition 40 normal gastric tissue were used as the control. Then steps the rest procedures according to the instruction of kit, the dilutions of antibodies both were 1:50.First, we dropped the first antibody and second antibody into slices, then washed the slices after 5 minutes, then stained with DAB, last, stained.again. We use The positive gastric adenocarcinoma tissue sections knowning of phosphorylation of FAK and Paxillin- as positive control and negative control with phosphate buffered solution(PBS). The datas were analysed by statistical software SPSS 13.0:Chi-squared test and Spearman's rank correlation were performed to compare difference bewteen groups and relativity analysis respectively. The significant difference was considered when the P value was less than 0.05.Results1 P-FAK expresses mainly in the cytoplasm and the surface of tumor-infiltrating.The positive signals of Paxillin staining express in the cytoplasm or cell membrane. 2. The positire expression of P-FAK protein was 47% in gastric adenocarcinoma tissues,and was higher significantly than those in normal tissues(5%,P<0.05).P-FAK expression was related to the cell differentiation,infiltration depth,lymph node metastasis, and TNM staging(P<0.05).3 The expression of Paxillin protein was 70% in gastric adenocarcinoma tissues,which significantly was higher than those in normal tissues (30%,P<0.05). It was observed that Paxillin expressions were related to the differentiation and the depth of gastric adenocarcinoma tissues and lymph node metastasis, and TNM staging(P<0.05).4. There was a positive correlation between the expression of P-FAK and Paxillin in gastric adenocarcinoma (r=0.505, P<0.05)Conclusions:1. The expression of phosphorylation of FAK is higher in gastric adenocarcinoma tissues than those in normal tissues. It is suggested that the higher expression of FAK phosphorylation is related to the ccurrence and development in gastric adenocarcinoma.2 It is indicated that the overexpressions of phosphorylation of FAK are related with the differentiation, infiltration depth, lymph node metastasis and TNM staging of gastric adenocarcinoma, suggested that its overexpression indicates cancer have higher invasive and transfer ability, it can be as judge of gastric adenocarcinoma aggressieness of reference index and prognosis.3 The overexpressions of Paxillin in gastric cancer tissues was significantly higher than that in normal tissues, which was correlated with cell differentiation, infiltration depth, lymph node metastasis and TNM staging,suggesting that Paxillin may be involved in the development of gastric cancer of biological behavior.4 Phosphorylation of FAK and Paxillin in gastric adenocarcinoma tissues were significantly positive correlated.It suggests that combinated expression of these two proteins have modulatory mechanism and involved in the regulation of many physiological functions of tumor cells.
