Aim:To study the effects of Huatanxiaoyu decoction on expressions of phosphatase and tensin homolog deleted on chromosome ten(PTEN), Focal adhesion kinase(FAK) and paxillin in rats with precancerous lesions of gastric cancer(PLGC), and to explore the related molecular mechanism.Methods:Ninety 4-5-week-old male SD rats were randomly divided into the blank control group(n=17) and the model group(n=73).except for blank control group, all other groups mainly used N-methyl-N’-nitro-N-nitrosoguanidine (MNNG), irregular diet and emotional stimuli to establish PLGC rat models. After successful modeling,15 rats were randomly selected from the blank control group, and 65 randomly selected were divided into the model group, the high dose Chinese medicine group, the medium dose Chinese medicine group, the low dose Chinese medicine group, and the Vitacoenzyme group,13 in each group, the model group were given normal saline, the treat group were given different dose Huatanxiaoyu decoction and the Vitacoenzyme.8 weeks later, to observe the pathological changes of gastric mucosa, and the expressions of PTEN, FAK and paxillin.Results:1.Theoretical research:Through access to the literatures of Traditional Chinese medicine and clinical medicine, to deeply understand the etiology, pathogenesis, diagnosis and treatment of PLGC, and to explore the theoretical basis and molecular mechanism of PLGC reversal of the Chinese medicine.2. The high dose Chinese medicine group significantly improved pathological conditions of gastric mucosa, and the improvement was better than the other treatment groups (P<0.05). The Vitacoenzyme group also improved PLGC changes(P<0.05) Compared with the model group. Compared with the blank control group, the expression of FAK and paxillin significantly increased (P< 0.01) and that of PTEN decreased (P< 0.05) in the model group. Compared with the model group, the expression of FAK and paxillin significantly decreased (P< 0.05) and that of PTEN increased (P< 0.01) in treatment groups. Compared with the vitacoenzyme group, the expression of FAK and paxillin increased more significantly in the high-dose Chinese medicine group (P< 0.05, P< 0.01).Conclusion:1. PLGC in traditional Chinese medicine "stomachache", "fullness", "spit acid" category. The disease causes machine are nothing but qi stagnation, cold coagulation, stagnation of heat, phlegm, blood stasis, and phlegm and blood stasis are common pathological stage of stomach long hair of the patients of PLGC. Modern medicine thinks, promoting blood circulation to remove blood stasis and resolving phlegm method can reduce gastric mucosal surface caused by phlegm and blood stasis factors, improve blood circulation, promote mucosal repair, by inhibiting oncogenes and (or) promotes the expression of tumor suppressor genes to regulate cell cycle and prevent PLGC to gastric cancer.2. Huatanxiaoyu decoction could clearly improve the pathological conditions of gastric mucosa to reverse the PLGC rats. The possible mechanism is to activate tumor suppressor gene PTEN, to regulate the dephosphorylation of FAK, and to down -regulate the paxillin through FAK/Src signaling pathway in aim to induce apoptosis.
