Background and Objective Hepatocellular carcinoma (HCC) that is one of the most common malignant tumors worldwide represents more than 4% of all cancer cases and has high morbidity and mortality。It causes at least 315,000 deaths each year, at the present time, Surgery is the most effective treatment。But because of its characteristic molecule biology and department of anatomy, only a fraction of the patients have the chance of Surgery。even though surgery,5-year survival rate is limited to 25% to 29%. After Surgery, No effective chemotherapeutic or other additional treatments are available。So the therapy of HCC is a cosmopolitan ticklish problem。From researchs,KISS-1 and COX-2 gene were a tumor-related genes KISS-1 was a metastasis suppressor gene of tumor, the gene encoding the major protein took its biological effects in tumor.Lots of studys were found that low expression or absence of KISS-1 was close to invasion, metastasis and poor prognosis.Example:breast cancer, esophageal cancer, gastric cancer colon cancer and so on. The COX was a rate-limiting enzyme during the synthesis of prostaglandins, which had two co-workers COX-1 and COX-2. COX-2, in the normal state, was generally not expressed only when cells were stimulated. In tumor cells, the cells were in a non-normal state, and COX-2 was highly expressed. Studys were found COX-2 in areas such as:stomach cancer, ovarian cancer, prostate cancer, lung tissue were highly expressed. It was closely related to the tumors.There were a few of reports about the expression of the KISS-1 and COX-2 in HCC in a large amount of literature at home and abroad, but the relativity study of KISS-1 and COX-2 was a vacuity and mostly researched the two genes from the level of protein but the relationship of the two genes with the development and transfer in HCC was obvious controversial。So the experiment will adopt RT-PCR to detect the expression levels of KISS-1and COX-2 in HCC group,liver cirrhosis (paracancerus) group and normal liver.To analyze what roles they played in HCC and the correlations of KISS-1,COX-2 to the infiltration and transfer of HCC.Samples and method:1. During May 2009-July 2010,39 cases of HCC,30 cases of liver cirrhosis and 10 cases of normal liver were collected from patients operated in general surgery of The Second Affiliated Hospital of Zhengzhou University who were not treated by radiotherapy,chemotherapy and other additional treatments before operation and had been confirmed by pathology2The experiment adopted RT-PCR to detect the expression levels of KISS-1 and COX-2 in HCC group,liver cirrhosis (paracancerus) group and normal liver.To analyze what roles they played in that normal liver developing into HCC and the correlations of KISS-1,COX-2 to the clinic pathologic parameters of HCC. The expression levels of KISS-1 and COX-2 in tumor group were analyzed by correlation analysis。3. The results were analyzed by SPSS13.0 statistical software package, Measurement data was used as X±s.Analysis of Variance,LSD-t and Pearson were used.a=0.05, a P-value less than 0.05 was considered significant.Results1.KISS-1 had expression in each sample; Its expression in HCC group were apparently lower than that of cirrhosis group and normal group (P<0.05).but there were no statically difference between cirrhosis group and normal group(P>0.05). 2.COX-2 had no expression in normal group;The expressions in HCC group and cirrhosis group were apparently higher than that of normal group (P<0.05), and the expressions in HCC group were also higher than those of cirrhosis group, but no significance (P>0.05)3.①The expression of KISS-1 were very low and had correlation with the level of AFP,envelop,ancerus thromus and Edmenson grade(p<0.05);But not correlation with age, sex,HBsAg and tumor size(P>0.05)。②The expression of COX-2 were very high but had no correlation with the clinic pathologic parameters of HCC(P>0.05)。4. The expression of KISS-1 were negatively related to that of COX-2 (r=-0.215, p<0.05).Conclusion1. KISS-1 could possible play an important role in HCC and had correlation with the capability of infiltration and transfer.2. The high expression of COX-2 was closely related to hepatocellular carcinoma, but had no relation with age,sex,tumor size,histological type,differentiation,metastasis,clinical stage and other clinicopathological parameters3.The expression levels of KISS-1 and COX-2 were negatively correlated.There was a mechanisms of feedback regulation each other in KISS-1 and COX-2.
