The topic determined the optimization compatibility of the proportion of icariin and PNS by cell culture method for promoting union of fracture, and prepared chitosan microspheres (PNS-ICA-CS-MS) by emulsion cross-linked. On the basis of single factor investigation, use the orthogonal experiments to optimized the best prescription in order to gain the best technics and prescription for perparing microspheres. And then study the shape characteristics, the release features in vitro, the preliminary stability, and the histocompatibility.The cell culture of osteoblasts Shows multiplier effect, the significant difference optimal fit is panax notoginseng-icariin(5:2). The PNS-ICA-CS-MS prepared according to the optimized technics and prescription have good dispersity and fluidity. Observed by SEM, the surface of spherical particles have pores. The size distribution of microspheres is narrow. The average particle size was about 63.71μm with 91.3% of the microspheres being in the range of 50~80μm. The average drug loading and encapsulation efficiency of panax notoginseng were(68.74±0.693)% and(16.32±0.155)% respectively, and that of icariin were(44.24±0.87)% and(3.3±0.161)%. In vitro release of panax notoginseng and icariin from microspheres could be described by Higuchi model and had a good sustained release characteristic. The microspheres have good Stability and biocompatibility.
