Icariin Stimulation On Expression Of OPG And RANKL Protein In Rat With Osteoporosis Fracture

Objectives: To observe the regulation of OPG and RANKL protein expression and the recovery of fractures by treating icariin with osteoporosis in rat.Methods: Forty eight female SD rats aged 4 months were divided into experimental group(n = 36)and sham operation group(n = 12)randomly.Sham-operation group only excision of a small amount of tissue around the ovary,the experimental group 36 were used bilateral ovariectomy in the manufacture of osteoporosis model,8 weeks after the determination of biochemical markers and bone mineral density,osteoporosis models were confirmed successful and then randomly divided into bone osteoporotic fracture(OPF)model group,icariin treated group and simvastatin positive control group.Each group was subdivided into 3 phase groups at the 4th,8th and 12 th week after fracture surgery.After successful modeling,the rat bone tissues were taken at three different time points respectively.The gross morphological changes of the rat bone were observed by HE staining.The expression of OPG protein and RANKLE protein in rat bone was detected by Western blot and immunohistochemistry.Results: Western blot and immunohistochemistry results showed that compared with the sham-operation group,the expression of OPG in the model group was significantly lower than that in the sham-operated group.After drug treatment,it was found that the icariin-treated group and simvastatin-positive the OPG expression in the control group at 4,8 and 12 weeks significantly increased.Compared with the sham operation group,the expression of RANKLE in the model group increased significantly,and the expression of RANKLE in icariin treated group and simvastatin positive control group was significantly decreased.Bone mineral density measurement showed that the treatment group of icariin than the model group in the three phases of bone mineral density was increased significantly,There were significant differences,It is say that icariin can effectively improve osteoporotic fracture rat femur BMD,accelerate fracture healing.Conclusion: Icariin can increase the expression of OPG,while inhibiting the expression of RANKLE,RANKL / OPG ratio changes,thereby promoting the healing of osteoporotic bone fractures.
