Objective:To study the effect of TTF1 to carcinoma blood vessel's growth. FFF1, 5,2',4'-trihydroxy-6,7,5'-trimethoxyflavone, is extracted from Sorbaria Sorbifolia ethyl acetate.Methods:TTF1 transplanted to the non-vascular region of chick embryo chorioallantocic membrane(CAM), and observed their effects of angiogenesis in CAM. 40 nude mice models with human hepatocellular carcinoma (HepG-2) were divided into 5 groups:negative group,5,7,4'-trihydroxy-flavone group, low TTF1 dose group, middle TTFl dose group and high TTF1 dose group,8 mice each group. After 21 days, nude mice models with human hepatocellular carcinoma were sacrificed. The expression of VEGF, KDR, b-FGF, HIF-1 and COX-2 mRNA and their protein in tumor tissues were detected with immunohistochemical method, Western blot and RT-PCR.Results:The micrangiums were growing well to be observed clearly in PBS group. Contrarily, they were obviously decreased in 25μg/ml TTF1 group and 50 g/ml group. The structure of micrangiums turned to vague and their colour became lighter to white.It was observed that, with the increasing does,TTF1 have a better suppressive function to angiogenesis. The results of RT-PCR and Western blot show that TTF1 can siganificantly down regulate gene VEGF, KDR, b-FGF, HIF-1 and COX-2 mRNA and their protein.TTFl have a suppressive function to carcinoma blood vessel's growth, and this mechanism is related with down regulation the expressions of VEGF,KDR,b-FGF,HIF-1 and COX-2 mRNA and their protein.Conclusion:Sorbaria Sorbifolia can siganificantly suppress the growth of hepatocellular carcinoma (HepG-2) cells.
