Objective:To study the role of TTF1-NP on the anti-inflammatory induced by LPS in human hepatocellular carcinoma cells.Method:MTT assay was used to detect the effect of different concentrations of LPS(0.1 mg/L,1 mg/L,10 mg/L and 100 mg/L)at different times(0h,12 h,24 h,48 h and 72 h)on the proliferation of human hepatoma HepG2 cells and selected the best point one;MTT assay was also used to detect the effect of different concentrations of TTF1-NP(5 ?pmol/L,10 ?mol/L and 20 pmol/L)on the proliferation of LPS(10 mg/L)induced human hepatoma HepG2 cells;the western blot assay was used to detect the effects of different concentrations of TTF1-NP on the expression of inflammatory cytokines IL-6,IL-8 and TLR4 proteins and the expression of Akt,p-Akt,mTOR,p-mTOR;the effects of TTF1-NP(10 ?mol/L)on the expression of IL-6,TLR4,and the phosphorylation of Akt and mTOR were detected after stimulated by insulin-activator of AKT/mTOR pathway.Result:1.MTT assay showed that different concentrations of LPS(0.1 mg/L,1 mg/L,10 mg/L,100mg/L)could promote the proliferation of HepG2 cells in a dose-time-dependent manner from 0.1 to 10 mg/L,0 to 48 h;2.MTT assay showed that different concentrations of TTF1-NP significantly inhibited the proliferation of human hepatoma HepG2 cells in a dose-dependent manner;3.Western blot analysis showed that different concentrations of TTF1-NP inhibited the expression of IL-6,IL-8 and TLR4-induced by LPS in a dose-dependent manner;4.LPS could promote the expression of p-Akt and p-mTOR,TTF1-NP significantly inhibited the expression of p-AKT and p-mTOR compared with the LPS group;5.TTF1-NP inhibited the expression of p-AKT and p-mTOR,and the expression of IL-6 and TLR4-induced by insulin.Conclusion:TTF1-NP inhibited inflammation responses induced by LPS in human hepatoma HepG2 cells,AKT/mTOR pathway play a key role.
