Clinical Study Of Neoadjuvant Chemotherapy Comparing Oxaliplatin Plus Docetaxel With Cisplatin Plus Docetaxel Before Radical Hysterectomy In Locally Advanced Cervical Cancer

Objective To compare the efficacy and toxicity of neoadjuvant chemotherapy with oxaliplatin plus docetaxel and cisplatin plus docetaxel followed by radical hysterectomy in patients with stageⅠB2-ⅡB locally advanced cervical cancer.Methods From February 2009 to December 2010,65 patients with stageⅠB2-ⅡB locally advanced cervical cancer were enrolled in the study. Scheduled for the patients were randomly divided into OT group (n=35) and PT Group (n=30). OT group patients received oxaliplatin 130mg/m2 plus docetaxel 75mg/m2, while those patients in the PT group were given the cisplatin 60mg/m2 plus docetaxel 75mg/m2. Both of the groups were managed with two cycles of neoadjuvant chemotherapy.Results (1) leukopenia, neutropenia in the experimental group were lower than those control group(P<0.05), thrombocytopenia and anemia did not differ significantly between the two groups(P> 0.05); (2) alopecia, nausea, vomiting and renal toxicity in the experimental group were significantly lower than those control group(P<0.01);(3) overall tumor response rate of drug and pathological response after neoadjuvant chemotherapy were similar in the two groups(P> 0.05); (4) diarrhea, liver toxicity and skin toxicity were similar in the two groups (P> 0.05);(5) peripheral nerve toxicity in the experimental group were significantly higher than those control group(P<0.01).Conclusions (1)The neoadjuvant chemotherapy with both OXA plus DTX and CDDP plus DTX have significant efficacy in the locally advanced cervical cancer,but the efficacy were not statistically different in the two groups; (2) The toxicities of OXA plus DTX were lower than CDDP plus DTX, the former would be significantly improving quality of life during neoadjuvant chemotherapy.
