Objective:Peri-implantitis is a kind of complication of implant prosthesis which often leads to the failure of dental implants. Porphyromonas gingivalis(P.g) is one main pathogenic bacteria of peri-implantitis. Gingipains K(Kgp) is a main toxic factor of P.g, which has a variety of biological activities and plays an important role in the bacterial adhesion. Because of its immunogenicity, it can serve as a kind of important vaccine antigens to prevent and treat peri-implantitis. This also can increase success rate of dental implants. This study aimed to construct pVAX1-Kgp as DNA vaccine against peri-implantitis, and observe its role after injecting the DNA vaccine into the animal model with peri-implantitis.Methods:1. Construction of the animal model (dog) with silk ligature.2. Construction of pVAXl-Kgp as a DNA vaccine. And then injecting the DNA vaccine into the animal by nasal drip, intramuscular injection and around salivary gland injections. Then collecting the samples, detecting the level of antibody IgG by ELISA in the blood. Results:Construction of pVAX1-Kgp was successful. The results of ELISA showed that IgG have a significant increase(P<0.05). Conclusion:The plasmid pVAX1-Kgp can increase the level of IgG and can be used to treat peri-implantitis.
