| Objective: Theoretically, explore the etiology and pathogenesis, therapeutic principle of threatened abortion and the research progress of the experimental methods of traditional Chinese drug-contained(TCD) serum and trophoblast by reviewing such kind of document research as threatened abortion, TCD serum, trophoblast, etc. Clinically, explore the clinical effectiveness of Anzi mixture by observing the improved situation of serum pregnancy hormone, clinical symptoms etc., before and after the treatment from the threatening miscarriage patients who are deficiency of spleen and kidney as well as stagnated-heat. Experimentally, explore the working mechanism of Anzi mixture on threatened abortion through inspecting the influence of TCD Serum in Anzi mixture on proliferation capacity of human trophoblast.Methods:include three parts:theoretical study, clinical research and experimental research.1. Theoretical study:illuminate the etiology, pathogenesis and treatment cognition of threatened abortion in the world history by Traditional Chinese medicine and Western Medicine two aspects; summarize the advantages and methods of TCD serum experimental research through the previous literature; summarize the recent research on the proliferation of human trophoblast from the field of cell biology. Based on these literature reviews, it puts forward the research thought and the topic basis of this task.2. Clinical research: observing 47 threatening miscarriage patients who are deficiency of spleen and kidney as well as stagnated-heat and divide them into 3 groups:Groups A(15 patients): Anzi mixture per os;Group B(19 patients):combination treatment of Anzi mixture per os and progestin muscle injection; Group C(13 patients):progestin muscle injection,4 weeks is one course of treatment. Observe the E2,P,β-HCG and the B ultrasonic before and after the treatment and mark the score of patients' symptoms.3. Experimental research:Take the serum from SD rats the stomach of which is poured with Anzi mixture and act on human trophoblast, find its influence on human trophoblast proliferation through MTT experimen. Results:In clinical research, success rate of pregnant keep is 93.3% in Group A,94.7% in Group B, and 84.6% in Group C; for the improvement of clinical symptoms, the three groups are all clearly improved in main symptoms(P<0.05), Group A and group B are all clear improved in secindary symptoms(P<0.01), and Group C is not improved in secondary symptoms(P>0.05); for pregnancy hormone growth, the three groups are all clearly increased(P<0.05), and Group A is obviously better than Group C in the growth of female hormone(P<0.05), and Group A,B and C share the same effect in the growth of P andβ-HCG. In the experimental research, TCD serum in Anzi mixture and Anzi mixture itself both play an obvious role in promoting the influence on the proliferation activity of human trophoblast. In the role of 24,48,72 hours'treatment, the influence of TCD serum in Anzi mixture on the trophoblast proliferation activity grows while its concentration grows; In the role of 48 hours treatment and 72 hours treatment, as long as the concentration reaches 15%, the influence become the strongest, but when over 15%, the influence reduce.Conclusion:Anzi mixture has the exact clinical effect on curing threatened abortion caused by deficiency of spleen and kidney as well as stagnated-heat, it can obviously improve the patients'symptoms as well as the level of E2,P,β-HCG in the early pregnant serum. TCD serum of Anzi mixture plays an obvious role in promoting human trophoblast proliferation. |
