Objective:Observe Heart-Protection Musk Pill (HMP) influence ventricular remodeling after Acute Myocardial Infarction (AMI) of model rats, by inhibiting the inflammatory reaction.Methods:Application of coronary artery ligation method and ligated the left anterior desecending coronary arteries(LAD) of rats to made AMI models, then observed the influence of HMP and simvastatin tablets to the ventricular remodeling of model rats,and the influence of interleukin-6 (IL-6), tumor necrosis factor-a (TNF-a), brain natriuretic hormone (BNP) in the plasma of rats and IL-6, TNF-a in the tissue.70 SD male rats were completely randomized, randomly selected 8 only as control group. Selected 10 rats to made sham operated models,8 rats of which were successful.The remaining 52 rats ligated the LAD to make AMI models,and 36 rats were successful, this 36 rats were randomly divided into four groups:model group, HMP equal dose group, HMP high dose group and simvastatin group, the number of rats in each group is 9. Control group,sham operated group and model group rats fed once a day, in AMI model manufacturing success 2 days after beginning, to the calculation of normal saline by 1ml/100g,1 times/day. HMP equal dose group, in AMI model manufacturing success 2 days after beginning, fed with HMP in solution 1ml/100g (per ml equivalent to the original drugs 1.4 mg),1 times/day. HMP high dose group, in AMI model manufacturing success 2 days after beginning, fed with HMP in solution 1ml/100g (per ml equivalent to the original drugs 2.8mg), 1 times/day. Simvastatin group, in AMI model manufacturing success 2 days after beginning, fed with Simvastatin solution 1ml/100g (per ml equivalent to the original drugs 0.2mg),1 times/day. Weighed each group once per week, then adjusted the dose of normal saline and drugs. Each group was fed to drawn. During the experimental process, all because of building model, and survival time after time based marketing requirements of the animals were excluded from the experimental observation of the project. After the success of modeling daily observation of the rats'mental health, hair, water intake, diet and activity.2 weeks after been fed, hemodynamic parameters of each experimental rats were measured, and used ELISA to determine TNF-α, IL-6, BNP in the plasma and TNF-α, IL-6 in myocardial tissue, weighted the left ventricular and calculated left ventricular mass index. the ischemia and non-ischemia myocardium area were be taken to make HE and Masson staining under light microscopy and immunohistochemistry of the inspection, and by transmission electron microscopy observation.RESULTS:(1) General lives circumstance observed:Compared with model group, equal and high dose of HMP,Simvastatin group all can improve the general lives of the AMI model of rats. (2) Hemodynamic test results:Compared with model group,the Hemodynamic test results of two dose of HMP and Simvastatin, can be improved significantly. (3) Changes of myocardial tissue pathology and ultrastructural:in two dose of HMP and Simvastatin rats, changes of myocardial tissue structure and the pathological got different degrees of improvement and content of collagen fiber in the myocardial tissue in the infracted and non-infracted areas have been reduced. They have significant difference when compared to model group. (4) Changes of LVW and LVWI:Compared with model group, in two dose of HMP and Simvastatin rats, LVW and LVWI of model rats have been obviously improved, and have significant difference. (5) High and equal dose of HMP, Simvastatin all can reduce TNF-a, IL-6, BNP in the plasma, TNF-a and IL-6 in myocardial tissue, they have significant difference compared to model group.Conclusion:HMP can improve the general lives circumstance and hemodynamic parameters, also can reduce the damage of myocardial cell histopathological and inhibit hyperplasia of collagen fiber of rats with AMI. HMP can Reduce the compensatory hyperplasia and hypertrophy of LV and reduce IL-6 and TNF-a in local myocardial and circulation of rats with AMI, inhibit inflammatory reaction occurs.This may be one mechanism of HMP improve ventricular remodeling in rats with AMI. Within the scope of the experimental dose of HMP, high dose of HMP have more obvious trend than the equal dose, in some experimental results.
