| ObjectiveTo observe the expression and of metastasis-associated protein (S100A4),matrix metalloproteinase 2 protein (MMP-2) and E-cadherin protein (E-cad) in Pancreatic Cancer, and to investigate the correlation and clinical significance of S100A4, MMP-2 and E-cad proteins expression.MethodsThe expression of S100A4, MMP-2 and E-cad proteins was detected by immunohistochemistry with SP method in 82 specimens of Pancreatic Cancer,43 specimens of and 21 normal tissue specimens. To explore the correlations with clinicopathological parameters among S100A4, MMP-2 and E-cad proteins expression in Pancreatic Cancer, and to investigate the correlations among S100A4, MMP-2 and E-cad proteins expression in Pancreatic Cancer. Results1. The positive rate of S100A4 and MMP-2 protein in Pancreatic Cancer respectively was 69.51%(57/82),58.54%(48/82), The expression of S100A4 in normal pancreas and Insulinoma respectively was 0(0/21),18.60%(8/43),The expression of MMP-2 in normal pancreas and Insulinoma respectively was 4.76% (1/21),13.95%(6/43),The expression of S100A4 and MMP-2 in the tissue of Pancreatic Cancer was significantly higher than that in Insulinoma and normal pancreas (P<0.05).2. The positive expression rate of E-cad protein in normal tissue specimens was 100%(21/21)and in Insulinoma was 72.09%(31/43), While in Pancreatic Cancer was 28.05%(23/82), The positive expression rate in Insulinoma and normal tissue specimens was much higher than that in Pancreatic Cancer (P<0.05).3. The expression of S100A4,MMP-2 and E-cad protein was associated with tumor size,lymphatic metastasis,TNM stages,pathological types and CA19-9 quantitative value (P<0.05); but not with age, sex, tumor site, smoking and diabetes(P>0.05),S100A4 and MMP-2 expressions were positively correlated in Pancreatic Cancer(r=0.626,P<0.05),Reduced E-cad expression was inversely correlated with S100A4 and MMP-2 expression in Pancreatic Cancer (the coefficient are r=-0.353 and r=-0.577,P<0.05).Conclusion1. The MMP-2 protein and S100A4 protein were highly expressed in Pancreatic Cancer. The E-cad protein was low expressed in Pancreatic Cancer. The expression of this three protein was associated with tumor size, lymphatic metastasis, TNM stages, pathological types and CA19-9 quantitative value, this indicated that high expression of the S100A4 protein and MMP-2 protein and low expression of E-cad protein in Pancreatic Cancer are related to the invasion and metastasis of Pancreatic Cancer.2. The correlation about S100A4,MMP-2 and E-cad protein in Pancreatic Cancer stated clearly that they were cooperation in the progression invasive and metastasis of the Pancreatic Cancer,The three proteins play a important role in tumor metastasis, Co-detection of the expression of S100A4, MMP-2 and E-cad proteins can be used to predict the metastatic potential of Pancreatic Cancer, assess malignant degree of tumour and can effectively evaluate the prognosis of patients with pancreatic cancer. |
