ObjectiveGliomas is the most common malignant tumor in the intracranial tumors. There is different degree of the peritumoral edema tissue surround gliomas. The prognosis of patients with malignant gliomas is very poor. It is difficult to eradicate by operation because of its invasiving growth. There is high recurrence rate after operation. The purpose of this study is to investigate expressions of MMP-2 and MMP-9 in peritumoral edema of different grades gliomas and their biological significance in tumorigenesis and progression of human gliomas.MethodsThe peritumoral edema of gliomas were taken from department of neurosurgery in First Affiliated Hospital of Zhengzhou University from July 2009 to November 2010, including 15males,9 females, mean age was 21 ages to63ages, the average age is 42 years old. Pathological grading of gliomas was classified according to the standard pathological grades for glioma.GradeⅠandⅡ14cases,gradeⅢandⅣ10cases. We combined gradeⅠand gradeⅡof glioma to low grade glioma(LGG),and gradeⅢand gradeⅣto high grade glioma(HGG). We take the of method of SP to detect the expression of MMP-2 and MMP-9 in peritumoral edema of gliomas and normal brain tissue. We used LSD-t tes,Bonferroni method to test the difference between normal brain tissue, low grade group and high grade group,and to analyze the relevance of MMP-2 and MMP-9 with Spearman level analysis.Results1. MMP-2: No positive staining of MMP-2 is found in the normal brain tissues. The positive staining was found in all of the 24 cases of the peritumoral edema tissues. The positive district average integral optical density of MMP-2 in normal brain tissues is 99.4163±5.21045.The positive district average integral optical density of MMP-2 in the peritumoral edema tissues of low grade gliomas is 135.9530±13.25774 and of high grade gliomas is 147.7253±10.22992.There is significant difference in MMP-2 expression between the peritumoral edema tissues of low grade gliomas and normal brain tissues(p<0.01); There is significant difference in MMP-2 expression between the peritumoral edema tissues of low grade gliomas and high grade gliomas (p<0 .01).2. MMP-9:No positive staining of MMP-9 is found in the normal brain tissues. The positive staining of MMP-9 was found in all of the 24 cases of the peritumoral edema tissues. The positive district average integral optical density of MMP-9 in normal brain tissues is101.9110±10.74317.The positive district average integral optical density of MMP-2 in the peritumoral edema tissues of low grade gliomas is 132.6771±8.23685 and of high grade gliomas is 150.5550±8.41266.There is significant difference in MMP-9 expression between the peritumoral edema tissues of low grade gliomas and normal brain tissues(p<0.01); There is significant difference in MMP-9 expression between the peritumoral edema tissues of low grade gliomas and high grade gliomas (p<0.01).Conclusion:1. MMP-2 in normal brain tissue did not express, and the expression of MMP2 in the peritumoral edema tissues of gliomas was positively correlated with the gliomas malignancy.2. MMP-9 in normal brain tissue did not express, and the expression of MMP9 in the peritumoral edema tissues of gliomas was positively correlated with the gliomas malignancy.3. The peritumoral edema tissues of gliomas play a certain role for glioma invasive growth. MMP-2 and MMP-9 can help to judge the invasion, malignancy and prognosis of gliomas, and become possible new directions in targeted therapy of gliomas.
