Objective:To analyze cerebrospinal fluid differences in protein patterns in rats after acute spinal cord injury by SELDI-TOF-MS technique and find significant changes differences in protein in CSF, To further explore the acute spinal cord injury repair mechanisms provide experimental evidence of proteomics aspects. Methods:Sixty male SD rats are randomly divided into 2 groups:high-dose methylprednisolone (MP) in the treatment group (A Group n=30) and Low-dose MP in the treatment group(B Group n=30). Prouduce SD rats model of acute spinal cord injury by Allen's method, intrathecal injection MP after Spinal Cord Injury:High-dose MP (0.075mg/100g) and Low-dose MP (0.001mg/100g). Cerebrospinal fluid will be extracted in 3h,6h,8h,12h. Detection of group A and group B of differentially expressed proteins in cerebrospinal fluid by SELDI-TOF-MS technology. And obtained spectrum of proteins in cerebrospinal fluid, filter out the differences between the proteins in each group by analysis package with ZUCI-PDAS. Results:Cerebrospinal fluid protein patterns were collected from 36 proteins,the peak M/Z is marked between 1000~20000. Four significant protein peaks are obtained between group A and group B.Conclusions:Application of SELDI-TOF-MS technique analysis of cerebrospinal fluid protein patterns in rats after acute spinal cord injury.There are significant differences in cerebrospinal fluid protein found between group A and group B.
