Background:Her-2 was a family member of the epidermal growth factor receptors. Her-2 expression positive was a special kind of gastric cancer, and showed a good effect in molecular targeted therapy. It is important for the treatment of gastric cancer to clarify the correlation between Her-2 gene expression and prognosis in gastric cancer.Aim:To clarify the relationships between Her-2 expression and prognosis in gastric cancer, and to demonstrate that Her-2 was an independent prognostic indicator for gastric cancer.Methods:78 gastric cancer patients were randomly selected according the inclusion and exclusion criteria, and the Her-2 gene expression was detected by Fluorescence in-situ hybridization (Fish). The correlation between Her-2 gene expression, gender, clinical characteristic, surgery, pathological data, tumor biomarkers, adjuvant chemotherapy, adjuvant radiotherapy, and prognosis were statistically analyzed. The primary endpoint was overall survival (OS). Survival curve was calculated by Kaplan Meier analysis. The difference of OS between Her-2 gene expression negative and positive was analyzed by the Log-rank test. The correlation between various factors and OS was analyzed by multivariate Cox regression analysis. Results:In 78 gastric cancer cases, the percent of Her2 gene expression positive was 15.4%. The median survival time of Her-2 negative patients was 744 days (95% CI: 525~963 days), whereas it was only 522 days(95% CI:175~869 days)in Her-2 positive patients (p=0.040). Multivariate Cox regression analysis showed that Her-2 gene expression was an independent prognostic indicator for gastric cancer (p=0.047) Moreover, operative pattern, margin condition, nerve invasion, position of lymph nodes metastasis, the effect of chemotherapy were significantly associated with OS (p<0.05). Conclusions:The prognosis of Her-2 positive gastric cancer patients was poorer than Her-2 negative gastric cancer patients, which had statistical significance (p=0.040). And Her-2 gene expression, operative pattern, margin condition, nerve invasion, position of lymph nodes metastasis, the effect of chemotherapy were independent prognostic factors (p<0.05).
