ObjectiveThis study is aimed to investigate the effect of Shensongyangxin capsule in heart failure rats with QT dispersion extension and connexin43 expression reduction induced by pressure overload.MethodsHeart failure rats models were builded by constricting abdominal aorta and randomized to Shensongyangxin capsule group, heart failure group or sham operation group. The ejection fraction was measured by cardiac color ultrasonography 8 weeks late. Shensongyangxin capsule group were lavaged with Shensongyangxin capsule for 8 weeks, heart failure group and sham operation group were lavaged with normal saline for 8 weeks. Measuring QT dispersion by inserting home-made electrode into subcutaneous. Mean arterial blood pressure was observed by left common carotid artery intubation. Rat left ventricular sections were fixed with 10% formalin, embedded with paraffin and cut into slices. Observing left ventricular morphostructure by HE staining, myocardial fibrosis by Masson staining and Connexin43 expression by immunohistochemicalstaining.Results1.8 weeks after surgery echocardiography show:Ejection fraction of heart failure group and Shensongyangxin capsule group were≤50% and significantly lower than sham operation group (P<0.05) but Shensongyangxin capsule group and heart failure group were not significantly different (P>0.05).2.8 weeks after treatment cardiac electrophysiology show:QT dispersion of heart failure group were significantly shorter than sham operation group (P<0.05), QT dispersion of Shensongyangxin capsule group were significantly shorter than heart failure group (P<0.05).3.8 weeks after treatment hemodynamic show:Mean arterial blood pressure of heart failure group and Shensongyangxin capsule group were significantly higher than sham operation group (P<0.05), Shensongyangxin capsule group and heart failure group were not significantly different (P>0.05).4.8 weeks after treatment HE stain show:Cardiomyocytes of sham operation group were aligned, heart failure group were misaligned, Shensongyangxin capsule group were more aligned than heart failure group.5.8 weeks after treatment Masson stain show:Myocardial fibrosis area of heart failure group were significantly increased than sham operation group (P<0.05), myocardial fibrosis area of Shensongyangxin capsule group were significantly decreased than heart failure group (P<0.05).6.8 weeks after treatment immunohistochemicalstaining show: Connexin43 expression of heart failure group were significantly decreased than sham operation group (P<0.05), connexin43 expression of Shensongyangxin capsule group were significantly increased than heart failure group (P<0.05).ConclusionRats with heart failure have prolonged QT dispersion, increased myocardial interstitial fibrosis deposition and decreased myocardial connexin43 expression. Shensongyangxin capsule can shorten QT dispersion of heart failure Rats, decrease myocardial fibrosis area and increase Connexin43 expression in myocardium. The data of this study suggested that the anti-arrhythmic effects of Shensongyangxin capsule may be related with the improvement of QT dispersion, the mechanism may be anti-fibrosis deposition function and improving the cardiac muscle connexin43 expression.
