| Fruiting bodies of Hypsizigus marmoreus (Peck) Bigelow have aesthetic form, closetexture, delicious taste, and special crab smell. It was called 'good smell in Tricholormamastsutake, good taste in Hypsizigus marmoreus'. Usual eat could resist cancer, defensecancer, strengthen physical health, prevent aging and prolong life. The main content of this study include: To begin with, the nutritional physiology ofHypsizigus marmoreus was studied. What's more, the original species culture medium qualityoptimization was studied in wheat proportion and water content. And then, the effects ofdifferent combination of wheat bran, soya bean meal, sawdust and corn meal on growth ofHypsizigus marmoreus was studied, and then, the effects of spend Pleurotus eryngii substrateto the yield of Hypsizigus marmoreus were studied. The last one but not the least one,Quadratic regression orthogonal design was used to study the culture medium optimizationfor Hypsizigus marmoreus carbon and nitrogen combination of liquid cultivation, and L49(3)orthogonal design was used to study the culture medium optimization for Hypsizigusmarmoreus inorganic salts combination of liquid cultivation. Main results are as follows:1. For the purpose of Hypsizigus marmoreus nutritional physiology, the size of colonyand surface mycelium growth vigor were used to assess the effects of various carbon andnitrogen sources, inorganic salts, vitamins and plant hormones on mycelium growth ofHypsizigus marmoreus. The results show that Hypsizigus marmoreus mycelium could usemonosaccharides, disaccharides and polysaccharides, which were provided. Soluble starch, glucose and lactose were the top three best carbon sources for mycelium growth. Solublestarch was the most utilized, followed in order by glucose and lactose. All the organic andinorganic nitrogen sources investigated improved growth with beef peptone being the best, followed by Glu and beef extract paste. Magnesium sulfate, sodium chloride and potassiumchloride all have certain promotion on mycelium growth of Hypsizigus marmoreus (Peck)Bigelow,1.5g/L magnesium sulfate is the best one which promotes the mycelium growth ofHypsizigus marmoreus (Peck) Bigelow significantly. Nicotinic acid, folic acid, VB1and VB2promote the mycelium growth of Hypsizigus marmoreus (Peck) Bigelow,2.0mg/L folic acidis the best one which promotes the mycelium growth of Hypsizigus marmoreus (Peck)Bigelow significantly. IBA,NAA and KT all have prominent promotion on mycelium growth of Hypsizigus marmoreus (Peck) Bigelow,2mg/L IBA is the best one which promotes themycelium growth of Hypsizigus marmoreus (Peck) Bigelow significantly.2. Original species culture medium quality optimization was studied in wheat proportionand water content, the studies have shown that, mycelium of Hypsizigus marmoreus couldgrow among the wheat proportion of68%to100%. With the increase of wheat proportion, themycelial growth rate rose up significantly. The mycelial growth rate was maximized when thewheat proportion was100%. This Water content had a significant effect on mycelium growthof Hypsizigus marmoreus, when the proportion of sawdust and water was1:1.6, the maximalmycelium growth speed was3.1935mm/d.3. Experiments were performed to determine effects of supplementation of wheat bran, soyabean meal, sawdust and corn meal on growth of Hypsizigus marmoreus using quadraticregression orthogonal design. The results estimated that, the most suitable proportion forfruiting bodies growth of Hypsizigus marmoreus was confirmed when the concentration ofwheat bran, saw dust, sawdust and corn meal were set at10%,1.5%,5%and15%,respectively.4. Effect of spenting Pleurotus eryngii substrates to the culture medium on the growth ofHypsizigus marmoreus were studied, the result showed that when the percentage of Pleurotuseryngii substrates increased from10%to40%, the sackful time was shorted from43d to29d,which improved production effectively, but had no effect on its biological characteristics.5. Effect of spenting carbon and nitrogen sources to the liquid culture medium ofHypsizigus marmoreus were studied using quadratic regression orthogonal design, the resultsshowed that, the best combination of carbon and nitrogen sources was26.00g/L glucose,25g/L soluble starch,0.25g/L yeast extract and0.4g/L peptone. Effect of spenting inorganicsalts to the liquid culture medium of Hypsizigus marmoreus were studied L9(34) orthogonaldesign, the results showed that, the best combination of inorganic salt on the basis of the bestmedium of carbon and nitrogen sources was0.5g/L magnesium sulfate,0.15g/L ferroussulfate,0.03g/L manganese sulfate and0.075g/L zinc sulfate. |
