The kelp residue femented by Hypsizigus marmoreus and the activity of antitumor and antioxidant of its products were studied firstly through the engineering of liquid fermention, the results are as follows:1 Two species of Hypsizigus marmoreus 31# and JB06 were used to ferment on the kelp residue at four concentrations, through visual observation and comparison of the amount of polysaccharides,we geted a conclusion that 31# had a better fermentation effects on kelp residue at two concentrations:5% and 10%, the outputof the polysaccharides from the supernatant of fermention was higher 16.2% and 7.7% percent than from the supernatant of control of pure kelp residue solution in the same concentration, higher 12.0% and 6.0% than from the supernatant of JB06. So 31# is picked as the strain.2 The medium was optimized through the orthogonal test(3 factors and 3 levels), 31# used as the fermentation strain, the best medium formula was:kelp residue 7%, glucose 0.25%, beef extract powder 0.09%, the output of the polysaccharides was 3.68g/L, higher to 59.81% than the blank test only concluled 7% kelp residue.3 The anti-free radical and oxidation resistance capacity of polysaccharides were tested by using kits, when the concentration of polysaccharides was 0.5mg/ml, the inhibition rate of the hydroxyl radical of the blank polysaccharides,31# polysaccharides, JB06 polysaccharides were as follows:29.4%; 47.1%; 38.8%.the inhibition rate of 31# polysaccharides, JB06 polysaccharides increased by 17.7% and 9.4% compared with the blank; when the concentration of polysaccharides was 10mg/ml, the anti-inhibition rate of superoxide anion radical of the blank polysaccharides,31# polysaccharides,JB06 polysaccharides were 11.7%,26.3%,28.5%, the inhibition rate of 31# polysaccharides and JB06 polysaccharides increased by 14.6% and 16.8%; the total antioxidant capacities of the blank polysaccharides,31# polysaccharides, JB06 polysaccharides were:1.811,2.164,2.050 units/mg polysaccharides, the total antioxidant capacity of 31# polysaccharides,JB06 polysaccharides were higher than the blank polysaccharide, respectively, higher by 19.5% and 13.2%. This experiment proved that 31# had a better effect on using kelp residue once again.4 The vitro anti-tumor capacity of the polysaccharides and ethyl acetate extract were tested by MTT, the inhibition rates of 31# polysaccharides and ethyl acetate extract on gastric cancer cells SGC7901 were 63.71% and 68.80%, which increased by 22.78% and 15.08% compared with the blank polysaccharides and ethyl acetate extract. |
