Objective: To study the spawn isolation of the wild medicinal fungus Phellinus spp. To produce the mother seed, the secondary spawn and the culture spawn. To optimize the solid culture medium and the liquid fermentation medium; to select the best fermentation conditions; To study the artificial cultivation techniques of Phellinus linteus.Methods: Spawn isolation was optained through the fruitinging body tissue isolation. We use the Phellinus linteus Gy~20 as the representative strain in this fllowing research. The formula of solid culture medium and liquid fermentation medium were optimized by the test of L9(3) 4 orthogonal design. Selection the optimum liquid fermentation conditions, such as the rotation speed, temperature, inoculum size, medium volume, pH and fermentation time. Production the secondary spawn and culture spawn. Summarization the artificial cultivation techniques from four factors , such as temperature, humidity, light, ventilation, culture time.Results: First, in this study 11 wild spawn of Phellinus spp.were successfully isolated.Among them, nine were identified by this laboratory: between them 5 were Phellinus linteus, 4 were Phellinus baumii, and the other two are still to be identified.Second, corn flour comprehensive medium is the best culture medium for culture mother spawn of Phellinus linteus and the optimum formula are: glucose, corn flour, peptone, MgSO4, KH2PO4, agar. Concentrations of several important components were 20 g·L ~1, 20 g·L ~1, 1 g·L ~1, 0.5 g·L ~1, 2 g·L ~1, 20 g·L ~1.Third, the optimum formula liquid fermentation medium in 100 mL flask of Phellinus linteus are: glucose, corn flour, peptone, bran, MgSO4, KH2PO4. Concentrations of several important components were: 15 g·L ~1, 25 g·L ~1, 0.5 g·L ~1, 5 g·L ~1, 0.5 g·L ~1, 2 g·L ~1.And the optimum fermentation conditions are: temperature 28℃, liquid volume 75 mL, inoculum 10%, initial pH 6.0, rotation speed 190 r/min, fermentation cycle 13 d.Fourth, artificial cultivation of Phellinus linteus main factors: Hair fungus phase, mycelium culture temperature 25℃;bmedium water content 60 % ~70 %; culture time 35 d ~40 d; ventilation one time a day; without light, darkroom culture. Primordia formation stage, the differentiation temperature 20℃~25℃; relative humidity 80%~95%; culture time 14 d~21 d; ventilation 2 times a day; scattered light irradiation. Long mushroom phase, growth temperature 18℃~25℃; relative humidity 90% ~95%; culture time 30 d ~40 d, ventilation 2 times a day; scattered light irradiation.
