Abstract/Summary:
In this paper, the3α-HSD transgenic rice was used as material to detect thecapacity of degradation of steroidal substances, to investigate the change of starch,soluble protein of transgenic rice seeds in the filling stage and agronomic changes, andto analyze the differential proteomics of transgenic rice. The results were as following.1. Measured the degradation of steroid in the different growth stages of rice,theresults showed that the capacity of all the3a-HSD transgenic strains to degradesteroid substances were significantly higher than CK, the degradation rate of T5-5strain was highest (18.10%). The degradation rate was the highest in bootingstage,significantly higher than heading and maturity, the degradation rate of headingstage was the weakest.The degradation function was the strongest in the tillering stage(23.83%) and booting (25.84%) of T5-5.2. The expression of the3α-HSD protein in root was detected: T5generation wassignificantly higher than T7and T6generations. The expression of the3α-HSD proteinin tillering stage was significantly higher than that in booting and heading stage. Thetarget protein expressions of four strains were significantly higher than CK's.3α-HSDprotein expression in the roots of T5-3 strains was up to2.36mg/g.3. To researched the change of the grain starch and protein in the filling stage,itindicted that the starch content in the transgenic rice strain's seeds was different withthat in the CK, The starch content in addition to T6-6 and T7-1 was significantly higherthan CK, other strains were no significant difference than in the CK. The proteinchanges of T5-1, T5-6 were similar to that of CK.Finally the protein content changes ofother strains have various differences.4. Researched parts of agronomic trait, the results showed that they had varyingdegrees of variation. Flag leaf length and total grain number variation were notobvious. Plant height varied greatly.All the strains were a dwarf tendency. Many traitswere better than CK in the T7generation.5. The degradation ability of steroids of transgenic rice were investigated.Theresults showed that the ability of the T5-5strain was higher than that of CK andothers.There were22protein spots (20spots were identified by MALDI-ToF/MSanalysis) with differential expression between T5-5and CK by proteomics, in which2 protein spots in expression decreased (No.11and17), while others increased. Theseproteins involved in energy metabolism-related (9spots), defense response-relatedprotein (5spots), resistance-related protein (2spots), and with the actived methylcycle and metastasis-related protein (2spots).
