Glyphosate is a non-selective systemic herbicide. We transform rice with a synthetic modified EPSPs gene and developed stable transgenic single-copy glyphosate-resistant lines. Field test prove that transgenic lines can complete normal growth period and the main agronomic traits did not change obviously.Bidirectional promoters are sequences of transcription initiation elements shared between divergently transcribed neighboring gene pairs that initiate transcription in both directions. In transgenic practice, usually, multi-gene transformation is required. Thus, using bidirectional promoters will make vector-construct easier and reduce the chance of transgenic silencing caused by repeatedly using the same promoter. In this article, we did some bioinformatics' analysis on TIGR version 5 genes of Oryza sativa (japonica cultivar-group) genomic sequences that are annotated as expressed. According to previous study in human genome, which identify a bimodal distribution from the distance of divergent genes and prove that the small one is where the bidirectional promoters lies, we identified 461 candidate bidirectional promoters. We calculated the correlation coefficient from the expression profile data of different types of gene pairs based on our private database which constructed with Affymetrix GeneChip? Rice Genome Array, and find that bidirectional promoters show more correlation over other gene pairs. We preliminarily count the appearance frequency of several cis-element and find that more than half of the candidate bidirectional promoters come with TATA Box on one stand only, which is significant lower than other promoters.Plant development is a synergic process composed of different genes that orderly express spatial and temporal. Gene expression regulation is a multilevel complex process, transcriptional level regulation is the most critical link. Promoter, as an important regulatory elements in transcriptional level, has become one of the hot spots in molecular biology research. With the spreading of genetic engineering, constitutive promoters are exposing issues, thus tissue or organ specific inducible promoters are chosen for a better regulation of plant gene expression.For a long time, cis-element study treat each single nucleotides as individual units, without considering the dependent among them. As we do not know which nucleotides are acting with a certain trans-factor when doing predicting, to merge a proper length of continuous bases together will be a change for considering this dependent. And this is what actually happens. Merging will also lead to an increase on the information content, which will make some more complicate analyses available without changing the current mathematic models.In this study, we transform continuous DNA sequence fragments to potential energy change when docking with certain trans-factors though physical properties, so that we can searching the mathematical law directly in the physical nature. However, as this subject is paused, no material results available so far.
