In this paper,60individual samples were collected from JingDe Xishan forestry station andYao Luoping national nature reserve. From the phenotype and molecular levels, Geneticdifferences were studied within and among the two Alnus trabeculosa populations, analysed thegenetic diversity by using variance analysis, RAPD marker technology and other methods.Theresults were as follows:1.Analysis of the phenotypic characteristics of60samples in the two natural populationsshows that Alnus trabeculosa has abundant phenotypic variation. leaf ratioï¹¥leaf widthï¹¥leafvenationï¹¥leaf length.2. The variation range of phenotypic differentiation coefficient is0.61%~16.67%. And theaverage coefficient of phenotypic differentiation is8.73%, while the average variation coefficientwithin population is91.27%.The variation within population is the major source. The diversitylevel within groups is higher than the diversity among groups.3.10primers were selected from40random primers which were sifted from InvitrogenCorporation in order to amplify polymorphic loci.In all,57DNA spectral bands were producted.Ithas51polymorphic locus, the polymorphic ratio is89.47%. The genetic diversity was judged byNei 's coefficient(0.2707), Shannon index (0.4137) and Gst index(0.0879). The geneticdiversity within the groups possessed91.21%. Gene flow(Nm)was5.1884, it was indicated thatthe genetic exchange level is high. Genetic distance between populations genetic distance was0.0703.4. Drawing of genetic affinity dendrogram of60samples of Alnus trabeculosa byUPGMA.In Jingde natural secondary forest,30individuals had close genetic relationship.But inYao Luoping natural population,the genetic relationship between30individuals were not as closeas the natural secondary forest in Jingde.Even that several were close to Jingde individuals. It justexactly explained that the diversity level within groups is higher than the diversity among groups.
