| By means of random amplified polymorphic DNA(RAPD)markers,genetic diversity and population genetic structure in 3 natural populations of Ph. nidularia were investigated in this study. Based on the research of population genetic structure strategies for the conservation of these germplasm resources of Ph. nidularia were also discussed. Seven samples,five forms of Ph. nidularia and two affined species were tested with RAPD in order to analysis their genetic variation. The results as follows:1 .A total of 90 individuals, representing 3 natural populations of Ph. nidularia in JiangSu and GuiZhou,were surveyed by use of 19 RAPD primers,which generated 307 reproducible and clear amplification products.out of 307,272 loci with molecular weight from 200 to 3000 were polymorphic and accounted 88.6%.Nei's gene diversity(h) and Shannon's Information Index(I) were 0.2948 and 0.5040 in the species level respectively.Individually,effective number of alleles(Ne) was 1.4872 and observed number of alleles(A) was 1.8860 in the species level. Coefficient of genetic differentiation (Gst) generated by Popgene32 was 0.8432. Genetic Identity and Genetic Distance between population were estimated, and the results indicates that geography distance influences the genetic differentiation.2.Using 19 RAPD primers,209 loci were amplified,and 146 of these were polymorphic(69.86%).The result indicated:(1)The Ph. nidularia could be distinctly distinguished from two affined species in the same genus.There was a genetic variation among five forms of Ph. nidularia.,(2)The classifications on bamboo under species based on RAPD markers is somewhat reliable. RAPD markers can be a powerful tool for genetic research on the infra-species of bamboo. |
