The mastitis not only damage dairy animals'lactating function and finally wash them out that caused the huge economic losses, but also the dietetic and physical health due to the serious impact on the quality of dairy products. Therefore mastitis of dairy animals is always one of the main topics of the disease research. In the relevant report of mastitis, the cow mastitis are in majority, with the incresing of dairy goats breeding scale and the number in the recent years, the mastitis not only affect the breeding, still seriously affected the output and the quality of milk and consumer's health. But so far, we still lack the epidemiology and pathogenic species survey in dairy goat mastitis system. In addition, the immune cells and molecular mechanism of the animal whose body be infected with the pathogenic from the breast also not been elucidated. All of these will inevitably affect the implementation of mastitis prevention and specific strategies. It is important to research the development of the dairy goat breeding and protecting the consumer's health.In this study, subclinical mastitis of 4 dairy goat feeding farms in Guanzhong was investigated using Lanzhou Mastitis Test (LMT), we collected some samples of milk which were positive in the way of asepsis. Filtered them through the organism, isolated and purified them, then identified out of the pathogenic bacteria and then serotyped them. Meanwhile the model of mastitis was successfully replicated used the isolated pathogenic bacteria and various of cytokines concentration in sera and milk in the occurring process of experimental mastitis had been detected. The following was the main research results:1. The prevalence rate of subclinical mastitis of 4 feeding farms were 31.30%, 66.00%, 32.30% and 42.60% respectively, the average prevalence rate is 39.52%. Four kinds of pathogens what were Staphylococcus aureus(S.aureus), Staphylococcus epidermidis, Escherichia coli(E.coli) and Krebs's bacteria were separated and identified. Among them escherichia coli O serum type contained mixed O117 / O107 and O117 type.2. In the experiment, goats of group E.coli were abnormal in all physiological indexes after infected 4 hours, but returned to normal after 72 hours; While the physiological indexes of group S.aureus changed obviously after intection, there was even death individually. Rectal temperature of goats showed an variation of first increase and then decrease, group E.coli and group S.aureus were respectively reached to peak after reperfusion at 4 hours and 6 hours, their temperature were still higher after infected 72hours than before infection. Compared with healthy goat, group S.aureus breast biopsy revealed that there were a large number of cells infiltrated in acinar chamber.3. In experimental mastitis, the cytokines of E.coli group were first rose and than declined, got a peak at 24h and in S.aureus group they were gotten rising . The cytokines concentration of E.coli group in the serum attained the peak totally within 24h after infection; But in S.aureus group the cytokines concentration were first rose and than declined and attained the peak totally at 4h excepted to the IL-2, IL-2 was gotten declined.
