Cow Mastitis is a High incidence of a disease in Dairy cow Cultivation Industry.After infected with Pathogenic Bacterium,cytokine was released to regulate the immune response.Different cytokines may induce or inhibit apoptosis in different cell,the factors affecting Bovine Mastitis milk somatic cell apoptosis in several cells have not been reported.82cow milk samples come from a certain farms were pathogens isolated in this study,cytokine IL-6,IL-8,TNF-a levels were measured by using RIA,its’correlation were discussed.Milk somatic cells were counted by using DMSCC,correlation between IL-6,IL-8,TNF-a and changes in SCC were discussed. Milk somatic cell samples were isolated and prepared,flow cytometer analysis was used to detect apoptosis of milk somatic cells,and apoptosis rate in lymphocytes,macrophages, neutrophils,mammary epithelial cells were calculated,correlation between IL-6,IL-8, TNF-α and apoptosis rate of milk somatic cell were studied.The results show that: IL-6,TNF-α,IL-8in milk were arisen with the mastitis inflammation intensified;there has some correlation between IL-6,TNF-α,IL-8and SCC,and the correlation coefficient R were0.55953,0.58602,0.90170;the IL-6levels in case of Bovine Mastitis were significantly negatively correlated(P<0.01) with epithelial cells, macrophages,lymphocytes,and neutrophil apoptosis,and the correlation coefficient absolute value is close to1or up to1between IL-6and neutrophils,correlation between them were most obvious.This study shows that:the levels of IL-6,IL-8,TNF-a in cow milk and the number of somatic cells show a significant positive correlation,IL-6can inhibit apoptosis of various cells in milk,TNF-a can promote apoptosis of epithelial cells in milk.
