wheat stripe rust,caused by Puccinia striiformis f.sp. tritici, is one of the most serious diseases of wheat in the worldwide.The desieae has repeatedly pandemic in China and caused heavy losses in wheat production, it has threaten food safety science now.Research and production practice proves that planting resistant varieties are the most effective economic and security measures method in control of wheat stripe rust, However, due to the high variability of Puccinia striiformis f.sp. tritici, the limited breeding source, resulting the loss of stripe rust resistance in wheat. Geneticists and plant breeders has began to pay more attention to wild relatives and elite cultivars carring durable resistance in order to search new resistant germplasms.Seedlings of the parents, F1, F2 and BC1 progeny derived from the cross of M97(resistant)×Mingxian 169 (susceptible) were inoculated pathtypes Sun11-4, Sun11-11 and CYR29,CYR30,CYR31,CYR32,CYR33of wheat strip rust in greenhouse, the results of genetic analysis showed that1. After making M97 Inoculated into seven physiological races in china which could be infected by wheat stripe rust, on blades of M97 in seedling stage to CYR33 is 0~1, and the phenotype to other varieties of wheat is 0-0.The result showed that M97 manifested as immune or nearly immune for seven physiological races in china.This significant differences showed that M97 had good stripe rust resistance.2. The resistance to the Su11-4 and Su11-11 in M97 was controlled by one pair of dominant genes.3. Two pairs of independent dominant genes conferred to CYR31 and one dominant gene and one recessive gene indenpently conferred to CYR29,CYR30,CYR33.4. In segregating F2 population which was inoculated Su11-4, used SSR molecular markers technology,4 different SSR makers located on chromosome 1Dwere found including Xwmc222, barc229, Xwmc147andXwmc339, linked with the target gene (YrM97), with genetic distances of 3.9cM,4.8cM,7.6cMand11.2cM, respectively. The pedigree analysis and molecular maker results indicated the origin of the gene from Leymus mollis Trin and suggested that it was probably a new gene as YrM97 named temporally. The wheat cultivars from Huang-huai wheat grown area were detected by Xwmc222 and Xwmc147 as PCR primers showed that 9%of them have same location with Yr97,it provide the basis of utilize the new genes in wheat stripe rust resistant molecular marker-assisted breeding.
