| Catalpa bungei C.A. Mey is Bignoniaceae plant, which is a deciduous tree, has high value in wood-using and landscaping. In nature, Catalpa bungei has autogamous sterility phenomenon, result in few resources more and more. In this study, several Catalpa bungei tree's pollen and some floral organs were taken as subject, and optimization methods of pollen germination in vitro and pollen tube culture were studyed, additionaly, protein traits of different floral organs were also analyzed. The results were as follows:(1) The affect of different storage conditions on the Catalpa bungei pollen germination. The studies on the storage temperature have shown that the best Catalpa bungei pollen is in liquid nitrogen and -70℃Ultra low temperature freezer storage. The study On the thawing shows that the most appropriate way of keeping cryopreservation thaw is under the way in 35~38℃water bath and under the conditions of the thawing thaw 5min.(2) The affect of the training environment and culture medium composition on the Catalpa bungei pollen germination and pollen tube growth. The studies on the culture temperature have shown that the most suitable temperature ranges 24~28℃. The studies on medium pH show that Catalpa bungei pollen need training in weak acid environments, the optimum pH value was 5.0~5.6. Sucrose concentration of the culture solution studies show that sucrose concentration of 15g·L-1~25g·L-1 had a higher germination rate of pollen.On the PEG concentration in culture medium showed that the most suitable PEG concentration is 20g·L-1PEG-4000.(3) Catalpa bungei, in this experiment, several different floral organs protein content was determined, several proteins in different floral organs results showed that the protein content of different organs were changed regularly. All these show that the highest protein content in ovary, followed by the calyx, and style, or the minimum protein content of petals. In the result of the SDS-PAGE electrophoresis experiment, it shows that each type pollen protein molecular weight through the analysis concentrate in 100kD, approximately 20 egg leucorrhea locate quite evenly in each swimming lane from high, middle and low molecular weight region. The molecular weight is the 12.5kD protein for the different Catalpa bungei seed different organ's public protein, in addition, the various tree seeds' different organ also has its special expression protein.(4)This experiment has also carried on the determination to the different Catalpa bungei pollen protein content. It also shows that:the pollen protein content varies from the different Catalpa bungei seed. The electrophoretogram contrastive analysis reveals which carries on to three kinds of catalpa pollen soluble protein, the Yunnan mallotus japonicus and the Laoshan catalpa protein content high place concentrates about molecular weight 30kD, but Lianyungang catalpa calyx and big flowered pollen protein content high place about molecular weight 20kD. Finally, demonstrated by three kind of different catalpa pollen protein's IEF-PAGE electrophoresis result, three kind of catalpa pollen protein molecular weight between 24kD-135kD, however the protein isoelectric point mainly concentrates between pI4-7. Total may also obtain, the same molecular weight's protein possibly corresponds the different isoelectric point to distribute in each kind of pollen. |
