Tetracycline antibiotics is a kind of broad spectrum antibiotic, and can be used for prevention and treatment livestock disease or promote the growth of livestock and poultry feed additives. But the excessive use of tetracycline antibiotics can cause drug residues in the animal, or remain in eggs, milk, viscera organization, and eventually affect human health. In order to guarantee the cattle tetracycline drugs in standard is used and protecting human health and the healthy development of animal husbandry, so perfect residue monitoring system must be established.This experiment accordanced with the currently study high-performance liquid chromatography (HPLC) detection tetracycline antibiotic content pretreatment methods inspection process and reference government standard detection requirements in pork and eggs, established Ultra Performance Liquid Chromatography (UPLC) detection of pork and eggs tetracycline antibiotic content,according to consequence of two methods,confered to their volume of use dissolvent,separating degree, linear correlation, precision, accuracy rating and sensitivity, etc, to explore more update and precise applicable to the residue detection method of tetracycline antibiotics for veterinary drug residue monitoring system, and to provide scientific detection basis.The results were as follows: (1)The peak amplitude emerge sequence of HPLC detection pork tetracycline antibiotic hydroxytetracycline, ambramycin, chloroteracycline, retention time was 2.120 min,2.458 min,3.765 min.and in three of concentration 50μg·kg-1,100μg·kg-1,200μg·kg-1 their recovery hydroxytetracycline: 78.2%98.4%, The aberrance coefficients were 6.5%9.2% and 2.1%5.6%.ambramycin: 86.3%98.9% , The aberrance coefficients were 5.2%8.6% and 2.8%4.5%.chloroteracycline: 82.6%89.5%,The aberrance coefficients were 5.8%9.2% and 4.3%6.1%.The LOD all was 10μg·kg-1,the LOQ all was 20μg·kg-1.(2 )The peak amplitude emerge sequence of HPLC detection eggs tetracycline antibiotic hydroxytetracycline, ambramycin:, chloroteracycline, retention time was2.120 min,2.458 min,3.765 min.and in three of concentration 50μg·kg-1,100μg·kg-1,200μg·kg-1 their recovery hydroxytetracycline:71.5%83.4%, The aberrance coefficients were 3.8%8.9% and 2.8%4.5%. ambramycin:77.5%82.3%,The aberrance coefficients were 4.1%8.7% and 4.8%6.1%.chloroteracycline:71.7%79.5 , The aberrance coefficients were 3.1%7.6% and 3.9%5.8%.The LOD all was 25μg·kg-1,the LOQ all was 50μg·kg-1.(3) The peak amplitude emerge sequence of UPLC detection pork tetracycline antibiotic hydroxytetracycline, ambramycin, chloroteracycline, retention time was 0.581 min,0.864 min,1.552 min and in three of concentration 50μg·kg-1,100μg·kg-1,200μg·kg-1 their recovery hydroxytetracycline:96.8%103.5%, The aberrance coefficients were 3.8%7.3% and 2.4%4.2%. ambramycin: 95.4%102.2%, The aberrance coefficients were 3.9%8.1% and 2.7%5.9%.chloroteracycline: 94.4%98.5%, The aberrance coefficients were 4.5%6.9% and 2.2%4.6%.The LOD was hydroxytetracycline 0.001 ng·kg-1, Ambramycin 0.001 ng·kg-1. chloroteracycline 0.010ng·kg-1. the LOQ was hydroxytetracycline 0.25ng·kg-1,Ambramycin 0.25ng·kg-1, chloroteracycline 0.50 ng·kg-1.(4) The peak amplitude emerge sequence of UPLC detection pork tetracycline antibiotic hydroxytetracycline, ambramycin, chloroteracycline, retention time was 0.581 min,0.864 min,1.552 min and in three of concentration 50μg·kg-1,100μg·kg-1 , 200μg·kg-1 their recovery hydroxytetracycline: 82.5%99.8%, The aberrance coefficients were 2.9%7.5% and 2.3%4.8%. ambramycin: 82.5%94.9%,The aberrance coefficients were 2.6%7.1% and 2.8%5.1%.chloroteracycline: 83.0%97.0%,The aberrance coefficients were 2.1 %7.2% and 4.1 %6.2 %.The LOD was hydroxytetracycline 0.01 ng·kg-1,Ambramycin 0.01ng·kg-1,chloroteracycline 0.02ng·kg-1,the LOQ was hydroxytetracycline 2.5ng·kg-1,Ambramycin 2.5ng·kg-1, chloroteracycline 5.0 ng·kg-1.The study demonstrated that result of HPLC and UPLC analysis of tetracycline antibiotics in pork and eggs were both to coincidence government standard and the UPLC was a method that had shorter parsing time, higher accuracy, higher precision and higher sensitivity comparing with HPLC to detect the residue of tetracycline antibiotics in animal-derived food.
