Cancer, also called malignant neoplasm is a critical illness which serious threatened to the health of human life and quality of life. Experts predict by the year2020global population would be over8billion, and the cancer patients with new cases would reach20million,12million would die from cancer. Cancer will be the number one killer of the human health.Anthraquinones is a kind of very effective and commonly used antitumor drug. Since since the end of last century with doxorubicin, as a representative of the anthraquinone drugs in clinical application, still be applied to a wide range of solid tumor and leukemia chemotherapy. Clinical common anthraquinones chemotherapy drugs include doxorubicin, epirubicin, pirarubicin, aclacinomycin, daunorubicin, mitoxantrone and demethoxydaunor ubicin, etcEdible flavors as raw materials are very important to food industry production. They are used for increasing the food smell, improve texture, to improve and enhance the smell of food effect and the quality of food grade. Edible flavoring is the largest number of application in food, the most widely used kind of food additives, also is an important part of food additive.2-Hydroxy-4-methylbenzaldehyde as a kind of edible flavoring has been used widely in food industry, and it is a composite anthraquinones anti-cancer drugs important intermediate. So to study the synthetic process of2-Hydroxy-4-methylbenzaldehyde, to improve its industrial production process that has very important practical significance to the human life health and improving the quality of life. In this paper, including as follows:(1) m-cresol as raw material reacted with chloromethyl methyl ether to generate methoxy-(3-methyl) anisole in different solvents. The effects of different solvents, different ratio and reaction time were studied;(2) methoxy-(3-methyl) anisole reacted with tertbutyllithium to generate material A(a salt), then the dimethylform was joined to react,so we got the4-methyl-2(methoxy) methoxy benzaldehyde. The effects of different temperature, different ratio and reaction time were studied;(3)4-methyl-2(methoxy) methoxy benzaldehyde reacted with hydrochloric acid, take off protecting groups to generate2-Hydroxy-4-methylbenzaldehyde. The effects of different temperature, different reaction times were studied.The solvent effects of heavy crystallization were studied;(4) m-cresol and paraformaldehyde as raw materials which reacted to generate2-Hydroxy-4-methylbenzaldehyde. The different catalysts have been studied.Through the above study an optimal route has been given:m-cresol reacted with chloromethyl methyl ether to generate material A.Material A reacted with tertbutyllithium to generate material B. Material B reacted with dimethylform to generate material C. Material C reacted with hydrochloric acid to generate2-Hydroxy-4-methylbenzaldehyde.Then petroleum ether and ethyl acetate heavy crystallization get pure product.
