Lotu s seed skins,the spornioderm of the seed of NympHeaceaeNelumboAdans,have harsh to the taste with cool property.It was widelyused in traditional Chine se Medicine.It was used for clearing heat andpromoting diure sis and hot and humid purging dysentery with plantagoseed and Akebia Stem.In research,alkaloid s such as lien sinine,is01iensinine,nefefine and lotusine can better debase blood pres sure,antagonize arrhythmia,antagonize insulin resistance,etc.It has widetherapy foreground in desease of blood ve s sel in heart and brain.In thisthe si s,the chemical constituents and basic content of the lotu s seedskin s,that the polysaccharide s and alkaloid s from was extracted from thelotu s seed skin s,then is01ated,purified,and their antioxidant activity andstructure analysis were investigated.The detail s were showed below:The mixture from lotu s seed skins powder w-as extracted in threedifferent s01ution s.The chemical constituents of the lotu s seed skins vc-erethen preliminary studied with system chemical pretrial method.And bybasic methods,the basic ingredient s of lotu s skin content were studied.The results indicated that the content of crude protein,crude oil,carbohydrate,ash,crude fiber in lotu s seed skins were determined to be21 5%,3 1%,56 1%,5 5%,4 5%,in addition,there were still pilenols,tannins,saponin s,anthraquinone,navonoid,alkaloid in it.Ultrasonic-as sistanted extraction of Polysaccharide from lotu sseed skin s was inve stigated.By examining the effects of liquid/materialratio,extraction time,ultrasonic power,extraction times onPolysaccharide field using single~actor method coupled with orthogonalarray design.Re sults showed that the optimal value s param eters weredetermined as fellow-s:liquid/material ratio 30:1 (g/mL),extraction time1 5min,ultrasonic power 60W,extraction times 4.Under SUCh conditions, the yield of polysaccharide was 17. 37%,the content of polysaccharidewas 63 75%.UltrasOnic.as si stanted extractiOn of alkalOid S frOm 10tuS seed skinswas Investigated.By USing orthogonal experiment,the influence s ofvariOU S parameters such as extraction time,supersoniC power,ethan01concentratiOn or liquid/material ratio on extraction efficienCY werestudied taking yield of avenanthramides as study index.At the same time,taking clearanCe ability for DPPH,02。·and·OH as study index,itsantiOxidation in vitro vcas studied.The optimized param eters were asf0110VCS:extraction time,30min;supersonic power,70W;ethan01concentratiOn,80%;and liquid/material ratiO,25:1.Theavenanthramides of 10tus seed skins had better antiOxidatiOn in vitrO,andit had strong capacity of scavenging DPPH,02。·and·OH.Thin layer chromatograpHy(TLC)and column chromatograpHy wereUSed to enrich and purify total alkaloid S from lotu S seed skin S,Threecompound S were isolated and their structure S were identified by NuclearMagnetiC ResOnance (NMR) and ultraviolet spectrometry asN-methylcoClaurine(I),Pronuciferine(II),Neferine(III).Compound I,II and 111were isolated from thi s plant for the first time....
