Liqixiaoying tablet,which is composed of Radix bupleuri,Pericarpium citri reticulatae.etc.,is a compounded preparation of Chinese medicine and a clinical experiential prescription of professor chen ruquan,who works in hubei university of Chinese medicine. It has the function of relieving liver stagnation and regulating vital energy.clinical application indicate that it has a good effect on the treatment of goiter of sputum cross-resistance,and it is welcomed after more than ten years of clinical use by patient,which is worth using widely. This subject is mainly studied the preparation process of the preparation,and it is based on Traditional Chinese Medicine theory, application of modern scientific technology and methods, the preparation process of the preparation was systematic experimental studied to ensure scientific and rational preparation process.Part one:The study of the process of Water Extraction1. The purification process of PWE (water extraction liquid):The content of total saponin, total polysaccharide and the amount of extract as markers, the using of ethanol precipitation, chitosan clarification technology, macroporous resin Purification about the effect of the purification of PWE among the current Chinese enterprises were investigated, and the macroporous resin adsorption technology to purify PWE was finally determined. The final purification of process conditions were:water extraction liquid was concentrated to a crude drug concentration of 0.25g·mL-1, loading sample into the D101 macroporous resin column by the rate of 2mL·min-1, after loading the volume of column-like 4 times,then elution with 1 column volume of distilled water, combining eluent with loading sample effluent as a common effluent, and then followed with 20% ethanol of 5 column volumes and 70% ethanol of 5 column volumes elution resin column, collecting eluent, vacuum recycling alcohol by decompression, getting the purification liquid. The sample effluent was concentrated until the crude drug concentration of 1.0g·mL-1, by adding amount of 95% ethanol for ethanol precipitation in order to the concentration of 80%ethanol,by ethanol precipitation, precipitated polysaccharides were collected after standing by 12h, and combined with purification liquid that was the purified water extraction liquid.3. The drying process of water Extraction liquid:selecting the spray drying process for needing a short time and having a high retention rate of ingredients, drying process as follows:The purified solution was concentrated to the relative density for.1.05～1.10 (60℃), into the liquid temperature 60-65℃, into fluid speed 40～50mL·min-1, into wind temperature for 170 175℃, out air temperature for 85～90℃, under the condition of 45HZ frequency atomizer spray drying.Part two:The study of the process of The ethanol extraction1. The process of alcohol extraction:Its ingredients of Pericarpium Citri Reticulatae, Tangerine leaf folium Gitri Reticulatae and Turmeric were extracted by alcohol. The content of hesperidin, the dry extract percent as markers, the ethanol concentration, the quantity of alcohol,extraction times, soaking time on the effect of ethanol extraction were investigated, to optimize the ethanol extraction process by Orthogonal. The optimum ethanol extraction conditions were:adding twice of the amount of 60% ethanol absorbed complement, adding 12 times the amount of 60% ethanol, using reflux extraction methods extract 3 times, each time 1.5h.2. The purification process of Alcohol extraction liquid:The content of hesperidin, the dry extract percent as markers, chitosan clarification of purification process and water-deposition after Alcohol extraction were investigated, and the final purification method is water-deposition after Alcohol extraction.
