Liqixiaoyin tablet,which is made from Citrus Veticulata Blanco,Bupleurum chinense DC,other seven kinds of herbs,is an effective angent for treating Ying disease by reinforcing Qi,nourishing Yin,smothing liver,release pressure.We have made an experimental study on its preparation and quality standards.1.Investigation of the preparationIn point of extraction of volite oil,we used oil's volumn as evaluating indexes and investigated the optimum preparing and refining condition by the orthogonal test method and parallel test.The optimum extracting condition was:with 8 times amount of water soaking the three herbs and boiling 6 hours.In point of inclusion of oil,we used yield of inclusion complex and ratio of inclusion as evaluating indexes and investigated the optimum preparing and refining condition by the orthogonal test method and parallel test.The optimum including condition was:addingβ-CD withβ-CD:oil=8:1(g/ml),in 60℃,agitating 3h with rate 900(r·min^-1).The selection of the extraction technology:Compared extraction by water and ethanol,the aurantiamarin's concetration of extraction by ethanol is higher.Using the orthogonal design,the alcohol degree,alcohol quantity,extraction time and extraction times are considered.The optimization of the extraction technology is: 70%ethanol 8 times capacity of material medicine,extracted 2 times,2.0h per time.The selection of the defecation technology:By comparison of the aurantiamarin's concetration and remove impurity rate of concentration deposition and chitosan defecation,concentration deposition can't remove the impurity perfectly.By estimated of chitosan solution concentration,drug solution concentration rate,the added quantity,deposition time,chitosan defecation technology are determined as:concentration rate(1:10),added 1.0%chitosan acetate acid solution to 3.5%(g/100g)at 60℃,deposited 24h,filtrated.The selection of the drying spray technology:excipients were selected in terms of aurantiamarin's concetration,hygroscpicity and yield rate of dried powder with several exipients added,the process parameters were optimized by experiment.The optimal processing conditions were obtained and validated as follows:relative densitive of liquid feedstock wes:1.15～1.18,the input speed was 45～55 mL·min-1,intake temperature was at 155～165℃,outtake temperature was at70～80℃,atomization frequency 45HZ.On the basis of above studies,we selected the mixture of microcrystalline celluclose(MCC)and starch as the excipients to reduce the hygroscopy and increase the fluidity of extracting powder.2.Study of quality standardBupleurum chinense DC.,curcuma wenyujin Y.H.Chen et C.Ling, Trichosanthes kirlouni Maxim,Raphanus sativus L was indetified by TLC.For the sake of assays of aurantiamarin,the new high performance liquid chromatography method were developed.The average recoery rates was 102.27%and RSD was 1.39%.Assay methods were validated in the reaserch of concert.On the basis of the above-mentioned quantitative analysis methods for aurantiamarin,the quality standard of Liqixiaoying tablet was drafted.
