Objective:This experiment researched on the expressions of protein of NGF,ERs,IL-2,IL-6 and AR in hypothalamus-spleen-gonad axis of the elderly macaques, which are the western Sichuan subspecies, and discuss the law of expressive changes of the materials mentiond above caused by sex hormone and gender differences of elderly macaques.Method:Using immunohistochemistry SABC method to research on the expressions and variations of distribution of the NGF,ERα,ERβ,IL-2,IL-6,ARof the hypothalamuses, spleens,ovaries,testicles of 12 macaques, young and elderly(The male and female are half and half). Results:(1) The protein of NGF,ERα,ERβ,IL-2,IL-6,AR is widely distributed in every nucleus of hypothalamus, mainly located in cell nucleus and protuberance, a little bit in endochylema and cytomembrane as well. Compared with young macaque, the elderly macaques'expression of NGF,ERα,ERβ,IL-2,AR showed a prominent downward trend, while the variation of IL-6 increased obviously. Thus we speculated that the cell immune level of elderly organism descended while the body fluid immune level ascended comparatively. (2) There is protein of NGF,ERα,ERβ,IL-2,IL-6,AR widely existed in spleen, of which has a large amount of positive products in its red marrow area and the central artery surrounding the white marrow, yet less in the peripheral region of noduli lymphatici lienis of the white marrow and periarterial lymphatic sheath This indicated that NGF,ERα,ERβ,IL-2,IL-6,AR take part in the spleen's function of body fluid immunoregulation. The six materials mentioned above have larger amount of expression and higher strength of expression in young macaques' spleens than in elderly ones', proving that the declination of sex hormone(including estrogen and androgen) reduces the expression to NGF,ERα,ERβ,IL-2,IL-6,AR. (3) The protein of NGF,ERaα,ERβ,IL-2,IL-6,AR is distributed both in ovary and testicle and is revolved in the process of which the reproductive system functions. The decreased content of NGF,ERα,ERβ,IL-2,ARand the increase content of IL-6 of the elderly macaque both suggest that sex hormone has significant effect of adjustment on the six materials above. (4) There are differences in the expressions of NGF,ERa,ERβ,IL-2,IL-6,AR in hypothalamus and spleen based on gender. There is a tendency that the strength of expression of protein and the positive area occupation ratio of the elderly female macaque are lower than that of male ones, assuming the reason is that androgen level has less effect on hypothalamus and spleen than estrogen level, or the estrogen level declined notably when female animals stepped into the stage of old age whereas male animals'gonad age later than females,and that androgen can be tansformed into extrogen under the effect of arylhydrocarbon hydroxylase, thus supplement the content of extrogen internal to some extent. (5) Coexpression areas of NGF,ERqβ,ERβ,IL-2,IL-6,AR may existed in hypothalamus, spleen,ovary and testicle, and they may affect one another to adjust their coexpression point.
